Neural stem cells(NSCs)have the potential of multiple differentiation and can differentiate into neurons,astrocytes,and oligodendrocytes.Among them,oligodendrocytes play an important role in the myelination of axons in the central nervous system.Their abnormal differentiation or loss can lead to demyelinating diseases such as multiple sclerosis,white matter dystrophy and traumatic spinal cord injury.The differentiation and maturation of oligodendrocytes is regulated by the internal factors and external factors such as inheritance and epigenetics.Growth factors,neurotrophic factors,transcription factors and microRNA all have different regulatory effects on the differentiation and maturation of oligodendrocytes.Recent studies have shown that long non-coding RNAs are involved in the differentiation and maturation of oligodendrocytes,but their roles and mechanisms in oligodendrocyte differentiation are not fully understood.In previous work,we performed single-cell transcriptome analysis of mouse lateral ventricle cells and discovered a new long noncoding RNA Lnc784.In this paper,we studied the expression and function of Lnc784.The coding capacity test showed that Lnc784 has no protein coding ability.The results of rt-qpcr and in-situ hybridization showed that Lnc784 was widely expressed in various brain regions of mice,and the expression of Lnc784 was different in each brain regions.Furthermore,Lnc784 is distributed both in the cytoplasm and the nucleus,indicating that it can play a role at both the transcriptional and posttranscriptional levels.Lnc784 is an antisense RNA of the TCF7L2 gene and may exert its biological function by regulating the expression of the target gene TCF7L2.After up-regulation of Lnc784 expression in neural stem cells,the expression level of its target gene TCF7L2 was relatively decreased,the expression of genes related to oligodendrocyte differentiation was decreased,and the number of oligodendrocytes was decreased.When Lnc784 was down-regulated in neural stem cells,the expression level of its target gene TCF7L2 was relatively increased,and the expression of genes related to oligodendrocyte differentiation was up-regulated,and the number of oligodendrocytes was significantly increased.The above results indicate that Lnc784 affects the differentiation of neural stem cells into oligodendrocytes by regulating the expression of the target gene TCF7L2.Our findings enrich people's understanding of theregulatory mechanisms of oligodendrocyte differentiation and myelination,providing a new target for human intervention in the treatment of demyelinating diseases and nerve injury in the central nervous system.
