The microbial pathogens invasion is one of the main factors restricting the high yield of crops.Plants percept biotic stresses through receptors in the plasma membrane or intracellular,which can trigger downstream signal passway and active immune responses.Calcium signaling as the second messenger of the plant triggered by stresses in the early stage and plays an important role in signal transmission.As a typical PAMP,it is clear that how FLS2 percepts flg22.However,how flg22 induce intracellular calcium concentration([Ca2+]i)remains unknown.In this study,Arabidopsis transgenic line which stably expression AQ was treated by flg22 and acquired by luminescence-based Ca2+ imaging system.We want to find mutants defect in flg22 induced [Ca2+]i elevations..Finally,we want to obtain specific components of the flg22-induced calcium signaling pathway.The current results we achieved as followed:1.Through image phenotypic screening,we finally obtain some candidates which defect in [Ca2+]i elevations treated by flg22;2.We have whole-genome sequencing of a candidate,and now we are mapping this mutant gene and it is be found on chromosome 4;3.As the physiological phenotypes of the mutants shown,the selected mutants were not sensitive to At Pep1 and flg22,indicating that the mutated gene plays a role in plant immune;4.We found that the flg22 induced expression of PR1 gene in the mutant was lower than WT by real-time fluorescence quantitative PCR.This study will promote research of plant disease resistance related dearly calcium signaling and the use of genetic engineering methods to improve the disease resistance of crops.It will play an important role in the further breeding of disease-resistant plants and agricultural production practice.
