Thirty individual samples of Alcippe morrisonia were collected from Guangdong Nanling National Nature Reserve(SN)and Guangdong Chebaling National Nature Reserve(SC)in the South of Nanling Mountains,and from Shangyou County in Jiangxi Province(NG)in the North of Nanling Mountains.The genetic diversity among Alcippe morrisonia populations of the three places was compared by analysing their mitochondrial genes and nuclear genes to research the effects of Nanling Mountains on the genetic diversity of Alcippe morrisonia populations.142 individual samples of Alcippe morrisonia were collected from Guangdong Province,Yunnan Province,Jiangxi Province,Guangxi Autonomous Region and Hainan Province,to determine the sex ratio of Alcippe morrisonia populations,the differentiation of sex ratio between breeding season and non-breeding season and among subspecies of Alcippe morrisonia.The main results were as follows:Through the analysis on genetic diversity of Alcippe morrisonia based on the mitochondrial molecular markers,the average haplotype diversity of the three regions was high,while the nucleotide and genetic diversity were low.The population mean genetic distances of Alcippe morrisonia among the three regions showed that there was no genetic differentiation(P>0.05).AMOVA results showed that the three populations didn't have clear genetic structure(Fst = 0.0272,P>0.05).NJ phylogenetic relationships and haplotype minimum span network showed that the three populations had no obvious genetic differentiation.Neutrality Test and Mismatch Analysis showed that Alcippe morrisonia from SN had experienced historical population expansion.These results demonstrated there was no genetic differentiation between Alcippe morrisonia populations of north and south Nanling Mountains.Through the analysis on genetic diversity of Alcippe morrisonia based on the nuclear gene molecular markers,the average haplotype diversity of the three populations was high,while the nucleotide and genetic diversity were low.The structure of population genetic analysis showed that there was obvious genetic differentiation between populations of NG and SC,and between populations of NG and SN(P=0).AMOVA results indicated that there were no significant genetic structures among the three populations.NJ phylogenetic relationships showed that there was no clear evolutionary branch,which indicated there was no distinct genetic differentiation among populations of SC,SN and NG.Neutrality Test and Mismatch Analysis showed that Alcippe morrisonia populations in Nanling Mountains maybe had experienced historical population expansion.It was indicated from above results that there was no genetic evolution between populations of north and south Nanling Mountains and no clear evolutionary diversity was generated.It was deduced that there was no genetic diversity of Alcippe morrisonia between North and South sides of Nanling MountainsAlcippe morrisonia populations were sexed by amplifying CHD gene sequences.The male/female ratio of the Alcippe morrisonia was 1: 1.254 and there was obvious difference among populations of Alcippe morrisonia hueti,Alcippe morrisonia fraterculus and Alcippe morrisonia schaefferi.There was no significant difference in the sex ratio between the breeding season and the non-breeding season,indicating that there was no significant change in the activity of the females and males during the breeding season and the non-breeding season.
