Cell fusion refers to the process of combining cells into asexual cells by means of artificial intervention in vitro under the condition of mediation and culture.Cell fusion not only provides powerful means for the research of nuclear quality,gene regulation,genetic complementation,oncology,gene location,and senescence control,but also has extensive application value in genetics,animal and plant distant hybridization breeding,developmental biology,immunology,medicine,food and agriculture,and has become a hybrid breeding and drug screening.The key technologies of monoclonal antibody preparation,mammalian cloning and anticancer vaccine research and development are also discussed.Cell fusion methods include biological fusion,chemical fusion and physical fusion.What is widely used now is the physical fusion method.In the physical fusion method,the electro fusion method is the most widely applied.Compared with other methods,electro fusion method has higher efficiency,higher reproducibility,easier operation,no poison to cells,easier to observe and videotape the process of cell fusion,which is suitable for instrument application and standard operation.On the basis of the analysis of the principle of cell electrofusion,this paper makes a complete set of design based on the characteristics of the cell fusion chip,and compares the domestic and foreign research status in this field.Each specific functional module of the design is analyzed and implemented.After realizing the functional circuit and testing,and after comparing the design specification,we used human chronic myeloid leukemia cell K562 to carry out cell fusion experiment.The experimental results show that the design of the experimental device successfully completed the whole cell fusion process,and achieved the desired design purpose.In this paper,the negative feedback of impedance measurement is introduced in the design of cell electrofusion system,and it tries to find the law between the process of cell electrofusion and the change of cell solution impedance,and then uses its inner connection to control the process of cell electro fusion more intelligently,and finally to reduce the labor intensity of the operator and improve the cell fusion rate.Meanwhile,in order to slow down the effect of unipolar pulse in the cell process on the oxidation of electrodes,a bipolar pulse circuit is designed and implemented.In a word,the embedded cell electro fusion system has greatly improved the user experience,and makes the system more intelligent and miniaturized.
