M1 matrix protein and M2 ion channel protein are encoded by the M gene of influenza A virus,and play an important function in virus infection process.Two new subtypes which named H17N10 and H18N11 have been recently discovered in bats.According to reports other subtypes of influenza A viruses including positive highest H9 subtype can be detected in bats.They are likely to recombinant if exist simultaneously in bats,and the potential harm of the recombinant virus cannot predict,therefore,this study attempts to explore the features after bat influenza viruses recombine with other subtype influenza virus.The recombinant chimeric bat influenza viruses were characterized by virus growth dynamics in vitro cells and pathogenicity test in vivo mice.The results indicated that influenza virus M gene which source of mammals(PR8 Bat17)compared with M gene source of avian influenza virus(A2093 / H9N2),can significantly enhance the capacity of chimeric bat influenza virus replication and pathogenicity both in MDCK cells and mic.While in DF1 cells originated from poultry cells,on the contrary,avian influenza virus(A2093/H9N2)M gene can enhance the capacity of chimeric bat influenza virus replication.All these results indicate that the M gene is probably as a key factor for chimeric bat influenza virus.In the second part,through western blot and IFA with the corresponding monoclonal antibody or polyclonal antibody,weidentifed the expression of the prokaryotic and the eukaryotic expression vectors,and the M1 and M2 proten expression of Bat and A2093 two subtype viruses.The results showed that the M1 protein of the two subtypes was consistent with the results of both the prokaryotic expression,eukaryotic expression and the virus infection,and the two types of M2 protein showed inconsistent results.This indicates that there is an immunogenicity difference between the M2 protein of the two types of influenza virus.Therefore we can distinguish between bat influenza virus and avian influenza virus by M2 antibody,and thus provide new way of thinking for a more comprehensive understanding differences of bats flu and other influenza viruses,and also provide the basis for further research on the influenza virus M proteins related biological mechanism.
