Objective 1.This study was conducted to explore whether mesenchymal stem cells(MSCs)combined with endothelial progenitor cells(EPCs)can culture into vascular-like tissues in vitro,and whether the combination of two stem cells is better than single-cell application.2.This study was conducted to explore whether basic fibroblast growth factor(b FGF)can promote the culture of vascular-like tissue by two kinds of stem cells.Methods 1.MSCs and EPCs derived from the bone marrow of 4-6 weeks old C57BL/6 mice were successively isolated,cultured and expanded.2.Polyglycolic acid(PGA)was tailored,sterilized and embedded in rat tail collagen.3.Experimental groups: EPCs group,MSCs group,combined group(EPCs+MSCs),b FGF group(EPCs+MSCs+b FGF).Placing each group of cells on the PGA scaffold to make into a cell-PGA compound,and cultured the compound in vitro.4.The compatibility of cells with PGA was observed using inverted microscope and HE staining after being cultured in vitro for one week.5.The cell-PGA compounds were cultured in vitro for 8 weeks,and then were embedded in paraffin.HE and immunohistochemical staining(CD34 and ?-SMA)were performed.The staining results were scored by pathologists.6.SPSS 20 analysis software was used to analyze the difference of immunohistochemical staining between the groups.Results 1.After one week of culture,a large number of cells were attached to the PGA scaffold fiber,and the cells were compatible with PGA.2.After 8 weeks of culture,CD34 staining was positive in the combined group,the EPCs group and the b FGF group,and the MSCs group was negative.The score of the combined group was significantly higher than that in the EPCs group.The difference was statistically significant(p < 0.01).There was no significant difference on the score between the combined group and the b FGF group(p > 0.05).3.After 8 weeks of culture,?-SMA staining was negative in all groups.Conclusion 1.MSCs combined with EPCs can culture into vascular-like tissue in vitro and are superior to single cell culture.2.b FGF has no obvious promoting effect on culture of vascular tissue by MSCs and EPCs in vitro.
