Nowadays,aging population has become increasingly serious followed by the increase of the incidence of disease of cardiovascular and cerebrovascular diseases.Cerebrovascular disease(also known as the stroke)is a serious human health hazard of common diseases,is also a common disease and frequently encountered disease in clinical neurologist with the very high incidence,morbidity and mortality.Statistics show that in our country,the cerebrovascular disease is still in the first or the second cause of death in the population of cities in recent years.The study found that cerebral infarction caused by supplying the reduction or interruption,although infarction occurred near the center of the irreversible necrosis,but the ion balance and organization structure of ischemic brain tissue around over a period of time will still be able to maintain the normal level.These groups can be restored if restore blood supply in time,which indicates that active prevention,active treatment of risk factors and effectively controlled cerebrovascular disease will play an important role in the elderly health and longevity that will improve the quality of life.Optical Coherence Tomography(OCT)is a novel kind of imaging technique,with its unique coherence technique and advantage of interferometry,no radiation,noninvasive,high resolution and high detection sensitivity,observe the biology beneath the surface of the organizational structure.OCT technology as a noninvasive diagnostic tool has become an important tool of biomedical imaging and plays an important role in eye disease diagnosis.According to the imaging theory is different,OCT technology can be divided into two categories:Time domain coherence tomography technology and Frequency domain optical coherence tomography technology.The traditional time domain OCT technology need the mechanical movement corresponding to different depth of sample with imaging the disadvantage of slow speed,but limited by the signal acquisition speed.In order to improve this defect,frequency-domain OCT technology arises at the historic moment,frequency-domain OCT technology detects interference spectrum of sample light and reference light,based on the advantages of traditional OCT technology and realize parallel sampling,greatly improves the detection speed problem and signal-to-noise ratio,frequency domain OCT gradually replace the time domain OCT into mainstream.This paper first introduces the significance detection of cerebral blood flow and development background of OCT technology,based on the research of the basic principle of OCT and analysis of the various performance indexes of the spectral OCT,design and build a spectral OCT system.Simply introduces the basic structure of spectral OCT system,starting from the interference light path,then introduces the main component of system,detailed analysis and design including interferometer,scanning system and signal acquisition system,and analyzes the acquisition signal processing steps of spectral OCT system:pattern noise elimination and dispersion compensation.Then using the spectral OCT system verified the compensatory mechanism of carotid artery occlusion.Using Doppler phase velocity detection method to calculate flow velocity of the contralateral carotid artery,the Doppler algorithm are introduced and proposed a new simple method of calculating Doppler Angle.Using spectral OCT system collected contralateral carotid artery velocity of three groups of mice before and after ligation,through the comparison of three groups of data,confirms the compensatory mechanism of organism.Finally,using spectral OCT system observed middle cerebral artery brain cortex branching capillaries during a compensation experiment.Ischemia of cortex arteries branch blood vessels was occlusion by drug ET-1,combining OMAG imaging technology detect cortex capillaries and observed blood perfusion status before,during and after drug injection.Using OCT system on drug testing of ET 1 expand the application field and application scope.
