Influenza is an acute respiratory infectious disease that can be transmitted by humans and animals caused by influenza viruses.The glycoprotein hemagglutinin(HA)on the surface of influenza viruses can specifically interact with sialic acid(SA)oligosaccharides receptor on the surface of host cells,which is the biological basis for the influenza viruses to infect the host and then replicate and spread.For many hosts,certain influenza strains(such as the H7N9 and H5N1 viruses)have high pathogenicity and high mortality,which seriously threaten the health of animals and humans.It is critical that sensitive and reliable detection of influenza viruses is an essential part of the prevention and control of influenza.Traditional detection techniques rely mainly on immunological and molecular biological methods,which are often cumbersome and time consuming and highly dependent on instruments and biological samples,and their application is limited.This study developed a new method for influenza viruses detection based on nanobiotechnology.The method of this paper does not depend on biological products such as antibodies and aptamers and is easy to operate and can be performed in real time.The detection method established in this study uses the principle of fluorescence resonance energy transfer(FRET)between quantum dots(donors)and gold nanoparticles(acceptors).The authors synthesized glycan-modified quantum dots and glycosylation-modified gold nanoparticles by simple and efficient synthetic route,and characterized their chemical structure and physical properties.Both nanomaterials have good water solubility and spectroscopic properties and can bind to HA proteins simultaneously in aqueous solution,triggering FRET that can be detected sensitively.The author further explored the establishment of the FRET system for detecting influenza virus.By simply mixing the glycan-modified quantum dots and glycan-modified gold nanoparticles and the influenza viruses in a buffer,FRET can be induced to achieve the purpose of detecting influenza viruses.The reliability of the system was verified by the H1N1 and H5N1 strains.The detection limits of the two strains were 0.17 and 0.008 HA Titer,respectively.the detection time was 10 min.In this work,we research and develop a method for analyzing and detecting influenza viruses.It amplifies the signal by energy resonance transfer between the quantum dots and the gold nanoparticles to detecting influenza viruses because of the glycan-functionalized nanoparticles.These studies shows that the application of the method developed from glycan-nanoparticles to analyse and detect influenza viruses can avoid the use of biological products,achieving rapid analysis and detection of influenza viruses.
