Human follicle stimulating hormone(hFSH)is a glycoprotein gonadotropin synthesized and secreted by human anterior pituitary basophils.Its family members include glucoprotein hormones such as luteinizing hormone,thyroid hormone and human chorionic gonadotropin.They all share the same alpha-subunit as human follicle stimulating hormone,while beta-subunit varies with specificity.Human follicle stimulating hormone plays an important role in the field of human reproduction.It is responsible for regulating a series of physiological processes related to human growth,development and puberty sexual maturity.In females,human follicle stimulating hormone can promote the proliferation and differentiation of follicular cells,promote ovarian growth and development,and ultimately stimulate the maturation of germ cells;in males,the role of human follicle stimulating hormone is to distribute in testicular seminiferous tubules and promote sperm formation.It can be seen that human follicle stimulating hormone is one of the most important hormones in both sexes.If the secretion of human follicle stimulating hormone is low in normal women,it will lead to abnormal ovulation,which will lead to a series of diseases such as irregular menstruation,follicular dysplasia,anovulation,and eventually lead to infertility.For follicle stimulating hormone supplements,they can be divided into three categories because of their different sources.The first category is FSH obtained by extracting hypothalamus and pituitary gland,referred to as pituitary FSH;the second category is FSH extracted from urine of climacteric or menopausal women,referred to as urinary FSH;the third category is the coding sequence of alpha and beta chains of human follicle stimulating hormone by recombinant gene technology.Column inserted into eukaryotic high expression vector pON1.By DNA sequence analysis,the deduced amino acid sequences of the genomic DNA sequences of the alpha and beta chains obtained after the intron sequence was removed are identical to the natural amino acid sequences of the alpha and beta chains of FSH.The high purity FSH obtained by suspension cell culture is called recombinant human follicle stimulating hormone(rhFSH).These three kinds of supplements have their own advantages and disadvantages,among which pituitary FSH has low production and high price,and can not be widely used in clinic;urinary FSH,due to its origin in different human bodies,has great individual differences and impurities,thus causing patients with greater side effects and prone to allergic reactions,has been plagued by doctors and patients;therefore,recombinant human follicle stimulating hormone with high purity and specificity.Strong sex and other advantages are increasingly favored by patients.At present,from the domestic demand for recombinant human follicle stimulating hormone,the products are mainly imported.The only domestic "Jinsaiheng" recombinant human follicle stimulating hormone was marketed in 2015,which broke the dependence of domestic patients on imported drugs,reduced the cost of medication,and the product quality was reliable,and established the Pharmacopoeia standard.However,from the point of view of production capacity,the current process for each batch of 30 L training scale,can not meet the growing market demand.Through the study of the culture technology of recombinant human follicle stimulating hormone cells,the scale of each batch of 30 L culture technology was enlarged to 200 L culture scale,the protein quantity was consistent before and after enlargement,the production capacity was improved,the market demand was met,the production cost was reduced,and the patients benefited.
