In the early formation of liver and pancreas in mammals,both are developed from the precursor cells of the same endoderm,and transdifferentiation can be achieved under certain conditions.In the liver injury,cells around the bile duct were detected to express lgr5,and lgr5-positive cells proliferated and differentiated into hepatocytes and biliary cells to supplement damaged liver cells,which repaired the damage and was not detected in adult liver.The expression of lgr5 is also true in the pancreas.At the same time,the expression of pdxl gene and insulin gene Insulin in pancreas development was also restricted to islet cells,and no expression was found in other pancreatic cells.There have been many reports on the expression of lgr5,pdxl and Insulin genes in adult liver and pancreas,but the expression characteristics of these three genes in the liver and pancreas have not been reported.Therefore,the cre-loxp system and transgenic mice were used to detect the lineage of lgr5,pdxl and Insulin in hepatobiliary and pancreas by drug-induced,frozen section and immunostaining by cre recombinase system initiated by specific promoters Igr5,pdxl and Insulin.feature.The results showed that the Insulin gene was expressed in islet cells of adult mouse pancreas,and no expression was detected in pancreatic acinar cells and pancreatic ductal cells.Hepatic parenchymal cells,hepatic duct cells and bile duct cells in the liver were not detected.Expression was detected;pdxl gene was expressed in adult mouse hepatocytes,hepatic duct cells and cholangiocarcinoma cells,and expression was also detected in acinar cells,islet cells and ductal cells of pancreas;lgr5 gene was found in adult mouse liver No expression was detected in hepatic parenchymal cells,hepatic ductal cells,and bile duct cells,and no expression was detected in acinar cells,islet cells,and ductal cells in the pancreas,and no lineage characteristics were observed.The above results clarify the lineage expression characteristics of lgr5,pdxl and Insulin genes in liver and pancreas development,and lay a foundation for the study of related functional genes in liver and pancreas development...
