Bats are the only flying mammals which are next to rodents in the distribution and population.At the same time,bats are natural hosts for a variety of new and recurrent zoonotic viruses.More than 200 viruses have been discovered in bat.There are 135 species of bats found in China,which belong to 7 families,33 genera,and mainly distributed in densely populated and humid climates aeras,such as Central China,South China and Southwest China.Our study and investigation showed that the bats are rich in species and carrying a variety of zoonotic viruses in Guangxi-Vietnal border region.However,only a few number of bat species distrbuted in Xinjiang,which also carried an obviously regional characteristics of virus.In order to further explore the background data of bat-borne viruses and the prevalence of viruses in Guangxi and Xinjiang province,and reveal the cross-species and cross-domain transmission of bat-associated viruses,the 268 bats of 11 species in Guangxi and 122 bats of one species in Xinjiang were analyzed with viral metagenomics;Bat small circular DNA virus were detected,and the full sequence were amplified and the genetic of key protein-coding genes were analyzed.All these results will help us to understand the cross-border transmission of bat viruses and prevent new viral diseases.And the results obtained in our experiments are as follows:1.By deep sequencing and bioinformatics analysis,49 viral families were found in 268 Guangxi bats in 2016,and the total virus reads accounting for 83.0%vertebrate virus,1.2%plant virus,0.4%insect virus and 15.5%phage,respectively.2.The contigs of four viruses were analzyed on genetic evolution,such as picobirnavirus,calicivirus,polyoma virus and hepacivirus,and results show that:(1)There may be two new picobirnavirus and calicivirus in Guangxi;(2)Polyomavirus exhibits co-evolutionary characteristics with the host.Hepacivirus has abundant genetic diversity in bats,and it may be a variation strain of hepacivirus.3.The six viruses of bats,such as hepadnavirus,bocavirus,group A rotavirus,astrovirus,rhabdovirus and coronavirus,were detected and their evolution were analyzed,our results show that:(1)The astrovirus of bats were universally carried in bats,which exists in each sampling point in Guangxi,and astrovirus was detected in several species of bat.(2)The Scotophilus kuhli has a complex virus background,carrying four viruses including astrovirus,rotavirus,rhabdovirus and coronavirus,no more than two viruses other bats;(3)The hepadnavirus strains found in this study were interregional transmission with those previously found in Zhejiang and Guizhou;The evolution of Bocavirus is related to the host species of bats;Rotavirus can be transmitted to humans or recombined with human rotavirus;Astroviruses can spread in different bats,and can also spread among bats and other animals(rodents and pigs);There are significant evolutionary differences between the rhabdovirus discovered in this study and the strains previously discovered and reported by our team.These differences are enough to make it a representative member of the new viral genus;Coronavirus has a certain species bias in bats.The strain CoV-FCRA23 is highly homologous with the newly discovered HKU2 or SADS CoV,which indicates that the pig industry in Fangchenggang area is also facing the threat of the virus.4.We amplified the complete genome sequence of a gemycircularvirus?50 circoviruses and 5 anellovirus from bat samples in Xinjiang;3 gemycircularvirus?3 circoviruses and 8 anellovirus were amplified complete sequence from bat samples in Guangxi.;Homology comparison and evolutionary analysis showed that these virus strains belonged to 2 new gemycircularvirus,3 new circoviruses and 4 new anellovirus.(1)Gemycircularvirus:It was found in Guangxi Hipposideros larvatus and Xinjiang Pipistrellus abramus that which expanded the host range of gemycircularvirus in bats.According to ICTV regulations,3 gemycircularvirus in Guangxi and one gemycircularvirus in Xinjiang may each be a new gemycircularvirus.(2)Circovirus:We found 50 circovirus in Xinjiang,which can be divided into two types according to different coding regions.Full sequence homology comparison showed that there was a new circovirus strain in Xinjiang,and the other strains were variants of the strain;We found 3 strains of circovirus in bats in Guangxi,which may belong to two new circoviruses.(3)Anellovirus:5 strains of anellovirus were detected in Xinjiang,which may belong to a new species.8 strains of anellovirus were detected in Guangxi,which may belong to 4 new species.The anellovirus found in this study is different from the reported anellovirus in genome structure.It contains only one ORF and may belong to an unclassified circular virus.Conclusion:(1)Viral metagenomics technology was used to analyze the community structure of bat,bome virus in the Guangxi,enrich the background data of the virus in the region.At the same time,we analyzed the genetic diversity of 10 viruses and the possibility of cross-species or cross-domain transmission of some viruses.(2)We amplified the complete genome sequence of a gemycircularvirus?50 circoviruses and 5 anellovirus from bat samples in Xinjiang;3 gemycircularvirus?3 circoviruses and 8 anellovirus were amplified complete sequence from bat samples in Guangxi.;Homology comparison and evolutionary analysis showed that these virus strains belonged to 2 new gemycircularvirus,3 new circoviruses and 4 new anellovirus...
