Sporidiobolus pararoseus is a class of yeasts that produce the ability to eject spores.Among them,the species of Sporobolomces and Sporidiobolus accumulate carotenoids such as torulene,red yeast and ?-carotene.Carotenoids have a variety of important biological functions and application values,among which torulene has anti-salt stress and other oxidative stress.Recent studies have found that Sporidiobolus pararoseus has anti-oxidant and anti-prostate cancer effects.In this experiment,yeast cells with different pigment components were used as materials to detect cell death,mitochondria number,apoptosis and intracellular ROS levels under salt stress and temperature stress,and to analyze carotenoids function in preventing apoptosis.The distribution of carotenoids on subcellular structures was studied.The main conclusions were as follows:(1)Carotenoids were distributed on the membrane structure of plasma membrane,Golgi apparatus,mitochondria,etc.,yellow ?-carotene was more abundant in plasma membrane and Golgi,torulene was distributed more in the mitochondria.It was speculated that torulene were located on the mitochondrial membrane,and the mitochondria are protected by scavenging ROS to avoid membrane potential changes,thereby avoiding apoptosis.(2)Salt stress induced yeast cell death and was positively correlated with salt concentration;compared with non-pigmented cells,cell death rate was significantly lower in carotenoid-containing cells under salt stress(1 mol/L Na Cl);At 1.5 mol/L,yeast cells rich in torulene and red yeast had the lowest cell death rate,suggesting that red pigment has stronger salt tolerance.(3)0.5 mol/L Na Cl salt stressing induced apoptosis of mitochondrial membrane potential induced by ROS,and decrease the number of mitochondria;under salt stress,carotenoid-containing cells are significantly reduced compared with non-pigmented cells.Pigments prevent yeast cell apoptosis induced by changes in mitochondrial membrane potential by clearing mitochondrial ROS.Yeast cells rich in torulene and red yeast are more mitochondria than ?-carotene-rich cells stable under salt stress.(4)Temperature stress induced systemic damage such as oxidative stress and protein inactivation,which induces yeast cell death and apoptosis.Carotenoids reduce intracellular ROS produced by temperature stress,but do not prevent apoptosis.
