Japanese encephalitis virus(JEV)is a neurotropic virus,a member of the family Flaviviridae,which causes neuroinflammation by invading the central nervous system.Japanese encephalitis(JE)is caused by the JEV,which is a zoonotic infectious disease that seriously impacts on public health,and the mortality rate of JE is about 25%.In the present study,we found that miRNAs in mouse brain and neuroblastoma cells were down-regulated during JEV infection by MicroRNA microarray and MicroRNA deep sequencing analysis.MiRDB and TargetScan analysis indicated that miR-466d-3p targets IL-1?.In vitro transfection of miR-466d-3p mimic and inhibitor was further confirmed the targeting relationship between miR-466d-3p and IL-1?.We constructed all the non-structural protein of JEV,and found that the expression of NS3 in cells can inhibit the expression of host miRNA.In summary,the NS3 of JEV can severely inhibit the expression of host miRNA,which can promote the occurrence of encephalitis. |
