The cell membrane is a complex structure composed of lipid rafts and a variety of proteins.It is a special existence that can be closely connected to the cytoskeleton and the external environment.Studying the distribution characteristics of cell membrane proteins plays a vital role in our research on cell membrane structure and function.As the simplest and most intuitive method,biological imaging has been widely used in the study of cells.With the development of fluorescence microscopy,single-molecule localized super-resolution imaging provides new information to solve these problems,and hopefully provides new ways to reveal many features of cell membranes.And the selection of suitable and excellent marking methods plays a vital role in imaging.1.Using aryl thioacetate and chloro(hetero)aromatic hydrocarbons as raw materials,a simple and effective palladium-catalyzed carbon-sulfur bond synthesis method was established.This method can be applied to a variety of substrates,and achieves an ideal aryl sulfide yield of up to 95%.2.According to the research group's research on cell membrane transfer protein,a tryptophan fluorescent probe used to study tryptophan transfer protein was developed with a yield of more than 60%per step;at the same time,65%per step With the above yield,an omeprazole fluorescent probe was synthesized for studying H~+/K~+ATPase protein on cell membrane.3.Using dSTORM single-molecule imaging technology,use synthetic gefitinib fluorescent probe to label EGFR protein on A549 living cell membrane.Since gefitinib fluorescent probe molecule is a small size structure,it can enter into molecules and molecules.All the gaps between them to better identify and label the target epitope.With the EGFR protein labeled with gefitinib fluorescent probe,the average number of cells per square micrometer is 739±73,the average cluster area is 0.246?m~2,and the average number of dots in the cluster is 3.33±0.51.Compared with the commonly used cetuximab and EGF ligand probes,this probe obtains higher quality dSTORM super-resolution images.
