Verticillium dahliae Kleb.is a soil-borne plant pathogenic fungus which is widely distributed around the world.It can cause wilting and death of nearly 200 plants.Due to its strong pathogenicity,wide range of hosts and formation of microsclerotia,there is no effective control measure at present.Comparative genomics studies of V.dahliae have shown that a large number of lineage-specific regions(LSRs)produced by chromosomal rearrangement are the main intrinsic driving force of genetic variation of pathogenicity and play an important role.Therefore,revealing the functions of these specific genes has important scientific significance and practical value.In this study,based on comparative genomics analysis of V.dahliae,the biological functions of three JR2 specific genes of V.dahliae were analyzed.The main conclusions were as follows:1.Three specific genes related to the pathogenicity of the JR2 strain were identified.Three specific genes of the JR2 strain(Chr1g07050,Chr2g00380,and Chr6g02370)were up-regulated during the infection process,and the toxicity of the knockout mutant was significantly reduced.However,the mutant can form structurally and functionally attached branches and invasive nails,and then,penetrate celluloid membranes,and successfully invade host tissues.2.Specific genes Chr1g07050 and Chr2g00380 regulate the oxidative stress of pathogenic fungi.We hold an experiment to test the response of mutants after hydrogen peroxide threat.In this experiment,the single knock-out mutants of Chr1g07050 and Chr2g00380 showed sensitivity to hydrogen peroxide,indicating that it plays an important role in the defense response when host plant answers oxygen stress generated.3.These three lineage-specific genes are not involved in the growth and development of pathogenic fungi.There are no significant changes in the mycelial growth,conidial production,microsclerotia and formation of the knockout mutant.But on the cellulose membrane,the area of black production has decreased.It shows that the deletion of these three genes has a certain effect on the accumulation of melanin.
