Singing insects refers to a kind of insects that can make sound,such as crickets and katydids of Orthoptera,mosquitoes and flies of Diptera and cicadas of Hemiptera,etc.The biodiversity of singing insects is very rich,especially the variety of singing insects of Ensifera.At present,the classification of singing insects of Ensifera from Zhejiang Province mainly relies on the traditional taxonomic method,and there are few molecular data and song data about singing insects of Ensifera from Zhejiang Province.This paper studies the biological diversity of singing insects of Ensifera from Zhejiang Province through traditional taxonomy,DNA barcoding and song classification.The main contents and results are as follows:1.Based on the specimens collected in the biological museum of East China Normal University(ECNU)and the specimens collected in the field,this paper dealt with the singing insects of Ensifera from Zhejiang Province.4 families,12 subfamilies,29 genera and 48 species of Grylloidea were identified.1 family,7 subfamilies,26 genera and 56 species of Tettigonioidea were identified.1 family,1 subfamily,1 genera and 1 species of Gryllotalpoidea were identified.The citations and distribution information of each species were listed.2.Mitochondrial COI gene fragments of singing insects of Ensifera from Zhejiang Province were used to construct the gene barcode database.Then the phylogenetic tree was constructed by Maximum likelihood method to explore the role of DNA barcoding in species classification and identification.(1)201 COI gene sequences from 4 families,12 subfamilies,27 genera and 45 species of Grylloidea were obtained by PCR amplification.231 COI gene sequences from 1 family,7 subfamilies,26 genera and 56 species of Tettigonioidea were obtained by PCR amplification.These sequences enriched the gene barcode database.(2)COI gene has a high efficiency of species identification,but it is unable to distinguish T.tylacantha,T.forceps and T.hibinonis of Truljalia from Zheiang Province,so it can be used as an auxiliary means for species classification and identification.3.We studied the temporal characteristics of the song of 32 species of crickets of Ensifera from Zhejiang Province,belonging to 4 families,10 subfamilies,and 21 genera.We also studied the temporal characteristics of the song of 20 species of katydids,belonging to 1 family,5 subfamilies and 10 genera.(1)There were significant differences in the chirp duration,chirp interval,the number of pulse in a chirp,pulse duration and pulse interval among species.The song is species specific.(2)The temporal characteristics of the song of crickets and katydids are relatively stable.In some cases,the song within a species show different degrees of differentiation in different geographical environments.
