Iridoid is a kind of very important and widely distributed plant metabolites,generally in the form of glycosides in nature.With a variety of biological activity,it is often as a biomarker for chemical classification,and is a main effective components of many medicinal herbs and its preparations.Iridoid is formed of five yuan for a ring of carbon and oxygen six-member heterocyclic single terpenoids.Due to the structure of iridoid glycoside is various,and content in the natural product difference is big,quite a n?mber of trace elements,so the chemical characterization encountered great difficulties.As a important technical means of chemical characterization of complex system liquid chromatography mass spectrometry technology more and more get the attention of people.From the perspective of the molecular structure of iridoid glycoside,it contains 1 characteristics of the structure of the iridoid skeleton,namely 1 dihydropyran ring cis connect 1 unit of cyclopentane class structure,and usually connect a glucose in C1 position.The different structure of the natural products of the iridoid mainly displays in the n?mber of functional groups,species,location and space conformation.According to the basic skeleton of iridoid glycoside(the parent nucleus structure),it can be divided into cyclopentene,cyclopentane and epoxy silane type equivalent components.With different mass spectrometry system component,the cracking way is different.With ion trap mass spectrometry and high-resolution level 4 pole orbit trap mass spectrometry in positive and negative ion mode of epoxy ether type,the mass spectrometry cracking behavior of iridoid glycoside(picroside I,picroside II,sesam?m glycosides,catalpa alcohol)was investigated.Through depth analysis,the four type epoxy ether iridoid cracking way of picroside I,picroside II,sesam?m glycosides,catalpa alcohol is inductived and is concluded the mass spectrometry pyrolysis regularity of iridoid glycoside of the epoxy ether type.On the basis of rhizoma coptidis medicinal materials of iridoid compounds respectively by using high performance liquid chromatography/ion trap mass spectrometry(HPLC/ITMS)and ultra performance liquid chromatography/high resolution orbit trap mass spectrometry(UPLC/MS)have been characterized,7 iridoid,found two iridoid glycoside has not been detected in the rhizoma picrorhizae.With ion trap mass spectrometry and high-resolution level 4 pole orbit trap mass spectrometry in positive and negative ion mode for crack inside terpene ene ether(privet glycosides,privet glycosides,gentian hard and swertia food glycosides)discusses the pyrolysis behavior of mass spectrometry,s?m up the crack inside the mass spectr?m decomposition rule of secoiridoid glycosides.On the basis,the secoiridoid glycosides of fructus ligustri lucidi medicinal materials of rich inside secoiridoid glycosides was chemical charactered using HPLC/ITMS and UPLC/MS respectively,found that one iridoid glycoside has not yet been detected in fructus ligustri lucidi.The iridoid(picroside I.picroside II as well as ferulic acid)in rhizoma picrorhizae medicinal herbs and its preparations-qing gan jiu wei wan were quantitative analysised.First.with picroside I.picroside II as an index for its extraction conditions are optimized,by using the orthogonal experiment/single factor respectively to investigate the optim?m extraction conditions of rhizoma picrohizae medicinal materials and its preparations,investigates the methodology on the basis,then establishes the quality control method of rhizoma picrohizae medicinal materials and their preparations-qing gan jiu wei wan.
