This research based on primary liver cancer have the exact effect of oleanolic acid,ursolic acid and ginsenoside Rg3 as the model drug,preparation and loading of three kinds of traditional Chinese medicine composition of OUR-NLC nanoparticles,the realization of the three component balance package and letter of release of nano drug delivery system.To explore the nanostructured lipid carrier NLC as the carrier of the multi-component nano drug delivery system encapsulation and release,and cell toxicity study,and in vitro.1.Process and prescription optimization of OUR-NLC drug loaded nanoparticlesIn diameter,encapsulation efficiency,drug loading rate as the index of evaluation,review the process was optimized by single factor,and further by central composite design and response surface methodology to optimize its formulation.The optimization results show that,with NLC as the carrier,CTAB as surfactant,a milky appearance was prepared by emulsification method OUR-NLC three drug loaded nanoparticles solution,the total package sealing rate and drug loading were(44.35 + 1.46)%and(8.99 + 0.10)%.2.Study on freeze drying of OUR-NLC drug loaded nanoparticlesA choice of five parallel experiments were conducted to compare the frozen desiccant,freeze dry effect,the results showed that,5%sucrose or 5%mannitol as cryoprotectant,white powder OUR-NLC three drug loading nanoparticles lyophilized powder appearance is loose,solubility of good.After the verification experiment,the results show that,with good reproducibility,the process is feasible.3.Research on characteristics of OUR-NLC drug loaded nanoparticleEffect of OUR-NLC three of drug loaded nanoparticles by transmission electron microscopy of the determination of the particle size,morphology,PDI and Zeta potential of Zetasizer Nano-ZS90 laser particle size analyzer.The results showed,SH dual drug loaded nanoparticles were round in shape,uniform particle size distribution,and the PDI values were 99.04 + 2.09nm and 0.227 + 0.001,72 + 1.25mV,Zeta potential.Using dynamic dialysis method,using pH7.4 PBS as the release medium,constant temperature of 37 DEG C to avoid light constant speed(100RPM)drug release experiments of OUR-NLC three of drug loaded nanoparticles stirring in vitro.The results showed,OUR-NLC three drug loaded nanoparticles in vitro drug release conformed to Higuchi equation,the fitting coefficients R2 were 0.988,0.985 and 0.976.The results showed that the drug loaded nanoparticles,OUR-NLC three has certain slow release function.Analysis of entrapment and release rule,small molecular weight,small volume of the OA and UA more easily encapsulated,and the release rate is greater than the large molecular weight,large volume Rg3.
