My master's degree thesis is divided into two parts,the first part is mainly to detecte the pesticide residues of the rice and evaluate the health risks of pesticides in it;the second part describes an analytical method of the interaction between pesticide and macromolecules based on high resolution mass spectrometry.The first part:In order to increase productivity and improve quality of food crops,a large number of highly efficient use of pesticides are widely used in agriculture.Unfortunately,not all farmers follow good agricultural practices,leading to an increasing concern about food safety.Consequently,it is urgent to establish a reasonable scheme of pesticide application,as a basis for guiding growers to plant.In the thesis,we investigated the types of chemical pesticides which are often used in rice fields and the main application of pesticide varieties which farmers in Hubei Province usually choose,then collect rice samples from Hubei Xiangyang City,plant in the planting base of Hubei Green Plant Protection Co.,Ltd.Nowadays,many pesticides are more polar and less volatile or thermally labile,therefore,LC-MS is replacing GC-MS methodologies as the method of choice for pesticide residue analyses.In this paper,we used solid phase extraction-liquid chromatography-tandem mass spectrometry,optimized process in each stage of experiment,then accurately detected the residues of 12 pesticides in the food crops.Moreover,compared them with maximum residue limits(MRLs)of pesticides in foods,we find the Yongyou 4149 rice is more in line with the requirements of green health food.Through evaluating the health risks of some pesticides,we finally establish a set of green rice planting scheme.We expect to promote this administration program,guarantee highly yield and volume of grain.The second part:Proteins are the material base in organisms and pay an inprotant role in the maintenance of life activities.The interaction analysis based on protein has attracted much attention.Those studies not only have an important significance for pharmacological research in the biological body,but also expect to provide theoretical basis and technical support for the research and development of new pesticides.The present paper describes a mass spectrometry analysis method,which can rapidly detect the interaction between protein and pesticide small molecule.By using high precision mass spectrometry can rapidly determinate the interaction between BSA protein,SOD enzyme and pesticides,and the combined process of inquiry.Results show that,the process of BSA binding of methyl parathion is different from SOD enzyme binding with DDVP and avermectin.Moreover,results reveal that each BSA bonded with maximum five Parathion-methyls,it would not interact with emamectin benzoate.SOD can bond with two DDVPs and one avermectin,there is no interaction between SOD and thifluzamide.Finally,we compare the proposed method with standard methods,and the reliability of the proposed method is supported.It owns preponderances in less sample consumption,high detection speed compared with traditional strategies.Therefore,the proposed method would own certain practical values.
