It was demonstrated that flavonoids could scavenge oxygen radicals,regulate blood glucose level,and ameliorate insulin resistant.Tartary buckwheat,as a food and traditional medicine,was abundant of nutrients and bioactive compounds,which had been widely used.And the processing of tartary buckwheat damaged the quality of flavonoids by traditional processing.What’s more,it was confirmed that flavonoids of tartary buckwheat could ameliorate insunlin resistant,while the mechanism was not known.The current study combined chemical assay,HPLC,and animals’ experiments to identifiy the funtion in vitro and vivo of tartary buckwheat(Chuanqiao#1).The aim of the study was to investigate the antioxidant activities of tartary buckwheat with heating treatment.And then,we determined whether tartary buckwheat extracts could improve glucose homeostasis and insulin resistance of skeletal muscle tissue.The main conclusions of this study were as follows:(1)The result indicated temperature reduced dissimilar on the content of phenolics,flavonoids,rutin and quercetin.There were not signifacatly effect on the content of phenolics,flavonoids and rutin less on 180℃.But at 220℃,the loss respectively33.5%,46.4%,91.3%.On the contrary,the content of quercetin was increased respectively 5 and 43.6 times,under180 and 220℃.(2)Flavonoids,which were rich in rutin and lower quercetin,were the major components of phenols in tartary buckwheat.Treament with high temperature resulted phenols,flavonoids and rutin decomposition and increased quercetin,reducing the value of tartary buckwheat.(3)According to the result of antioxidant capacity,there was little effect on the antioxidant capacity of AAC,DPPH free radical scavenging capacity and reducing power with treatment of temperature under 180℃,but lower at 220 ℃,especially AAC.(4)Inflammatory factors induced inflammation and inhibited insulin action to regulate glucose metabolism,interfered glucose homeostasis and insulin resistance.Flavonoids in tartary buckwheat could ameliorate insulin sensitivity to regulate glucose homeostasis and enhance absorption of glucose with improving the content of glucose toleranc,insulin toleranc and synthesis of muscular and hepatic glycogen.(5)Tartary buckwheat extracts ameliorated insulin resistance in Mac-CM induced mice by regulating IKKβ/IRS-1/PI3K/Akt signaling pathway.Inflammatory induced insulin resistance and prevented signal transduction along IKKγ/IRS-1/PI3K/Akt pathway,with activating IKKβ,then increased the serine phosphorylation,while inhibited the tyrosine phosphorylation of IRS-1,then weaken the downstream phosphorylation of Akt.While tartary buckwheat extracts inhibited IKKβ and facilitated insulin signaling transduction. |
