N-[(2-hydroxy-3-trimethyl ammonium)propyl] chitosan(HTCC)is one kind of chitosan derivatives.Compared with chitosan,HTCC has good water solubility and flocculability,so it can be potentially applied in the field of polymer, industry, textile and other relative applications. In this paper, several novel quaternary ammonium chitosan derivative and hydrogels were synthesized. It provides a theoretical basis for the study that these hydrogel and derivative can be applied in the area of hydrogel dressing and antibacterial agent. The main contents and conclusions are as followed:(1)Firstly,poly(amidoamine)(PAMAM)was synthesized by Michael addition reaction and amidation reaction, with ethylenediamine and methyl acrylate as the raw materials. Then the hydrogel was prepared with PAMAM and HTCC as the raw material, glutaraldehyde as the crosslinking agent. The Fourier transform infrared(FTIR)and scanning electron microscopy(SEM)were adopted to characterize the structure and morphology of the hydrogel. The FTIR results showed that the hydrogel was synthesized successfully. It also could be seen from the SEM images that there were pore structure inside the hydrogels. The swelling test of hydrogel proved that it had excellent swelling and pH-sensitive properties. The increasing content of PAMAM or degree of quaternization of quaternary ammonium chitosan leaded to the increase of swelling property. With the increase of concentration of crosslinking agent and concentration of HTCC,the swelling degree decreased. The antibacterial results indicated that with the increase of PAMAM content, concentration of quaternary ammonium chitosan or concentration of cross-linking agent, the hydrogels had shown better antibacterial activities both against the Staphylococcus aureus(S.aureus)and Escherichia coli(E.coli).(2)A novel quaternary ammonium chitosan hydrogel modified by was prepared by citric acid(CA)using 1-(3-dimethylaminopropyl)-3-ethylcarbodiimide hydrochloride(EDC)and N-hdroxysuccinimide(NHS)as the crosslinking agent. The FTIR and SEM were adopted to characterize the structure and morphology of the hydrogel. The results showed that hydrogels were prepared successfully. The pore structures of hydrogel were loose. The increasing content of CA and degree of quaternization of quaternary ammonium chitosan leaded to the increase of swelling degree. The swelling degree decreased with the increase of the crosslinking agent and concentration of HTCC.The antibacterial results indicated that with the increase of the concentration of quaternary ammonium chitosan, the hydrogels had shown poorer antibacterial activities both against the S.aureus and E.coli, with the increase of the degree of substitution, the antibacterial effect of derivatives against the bacteria increased(3)Firstly,quaternary ammonium chitosan derivative was synthesized by Michael addition reaction and amidation reaction, with quaternary ammonium chitosan, ethylenediamine and methyl acrylate as the raw materials.By controlling the generation and material feed ratio, different groups of products were prepared. The FTIR result showed that derivatives were synthesized successfully. The antibacterial results indicated that with the increase of the generation and amino content, the antibacterial activities against the S.aureus and E.coli increased.Meanwhile, with the increase of the degree of quaternization, the antibacterial effect of derivatives against the bacteria decreased.
