Starfish has a wide variety of structural types of chemical constituents and strong biological activity,which has become a hot research.In this paper,the chemical constituents and antioxidant activities of Asterina pectinifera and Solaster dawsoni verrill in the Yellow Sea and Bohai Sea area of Dalian were studied.The preparation method of asterosaponin was studied.Four solvents of 95% ethanol,methanol,ethyl acetate and pure water,were used to extract asterinidae saponin.It was found that the extraction effect of methanol was the best.By comparing the two extraction methods of ultrasonic extraction and flash crushing extraction as auxiliary extraction method and hot reflux extraction method,it is found that the flicker extraction method is the best in both extraction and hot reflux extraction.The effects of four single factors,such as flash extraction time,solvent concentration,solvent concentration,ratio of solid to liquid,heat reflux extraction time,on the extraction rate of total asterosaponin were investigated.Methanol solvent combined with scintillation was obtained.The optimum conditions for the preparation of petrel saponins by assisted heat reflux extraction were as follows: methanol concentration 100%,ratio of material to liquid 1: 50,flash crushing for 5 min,heating reflux time 1.5 h.The crude product of petrel saponins was prepared by the optimized extraction process(70% ethanol instead of methanol solvent considering solvent harm).The extract was extracted by petroleum ether,ethyl acetate and n-butanol,and three different polarity(from small to large)were obtained.Thirteen compounds were obtained through systematic separation and purification of petrel extract by various chromatographic separation and purification methods.Six of them were identified by means of physical and chemical constants and spectral data analysis.They were asterosaponin P1(3?,5?-diisobutyryl-6?-isopropyl-piperidine-2,4-dione),stearic acid,alanine,thymine and(25S)-5?-cholestane-3?,4?,6?,7?,8,15?,16?,26-octol,of which 3?,5?-diisobutyryl-6?-isopropyl –piperidine-2,4-dione and alanine were isolated from Asterina pectinifera for the first time.Study on chemical constituents of starfish in thesolaster dawsoni verrill,extracted by 70% ethanol combined with hot reflux method to flash smashing extraction method as auxiliary extraction of saponins on solaster dawsoni verrill.After three different polar solvent was petroleum ether,ethyl acetate and n-butanol extraction of asterosaponin,get three different polarity(increasing)of asterosaponin extract.The n-butanol and petroleum ether extract of saponin of combining a variety of chromatographic separation and purification method,the separation system,nine compounds were obtained.On the basis of physicochemical constants and spectral data analysis method for the identification of three species of them were compound Pa-1,cis-thyminenol and cyclic(ile-leu)were isolated for the first time from solaster dawsoni verrill.The free radical scavenging(free radical,hydroxyl radical and superoxide anion free radical)of the extracts of asterinidae and solaster dawsoni verrill were determined.The paper provided a theoretical basis for the development and utilization of starfish resources.
