Polymersomes are high molecular aggregation with hollow sphere structure which are self-assembled by amphiphilic block copolymer.Compared to liposomes,polymersomes have excellent stability and have attracted wide attention in the areas including drug delivery and gene therapy,etc.Generally,drugs are loaded during the process of polymersomes preparation,which have the problems of low efficiency and limit drug species.So another loaded route can be replaced.Polymersomes can be loaded content(protein,drugs,fluorescence)through electroporation after polymersomes formed.The specific research contents of this thesis are as follows:(1)The research of polymersomes preparation by different block copolymer and different methods.The process of polymersomes self-assembled influenced by the structure of amphiphilic molecules,preparation methods and environment factors.This research prepared polymer vesicles with different morphology,different sizes through film rehydration and solvent exchange methods.Polymersomes were characterized by different technologies.This research also utilized phosphotungstic acid and uranyl acetate solution staining for SEM characterization to instead of spraying gold methodologies.The results showed that SEM images were clear and three dimensional,and surface details were also maintained.(2)The research of electroporation medicine into polymersomes.For traditional loaded method,drugs are loaded during the process of self-assembly,which have the problem of low efficiency.Previous research proved that medicine can be loaded into polymersomes through electroporation.In order to discover loading process,reverse electroporation proved that the pores formed during the process of electroporation.The contents were loaded into polymersomes firstly and then electroporation.Electroporation medicine into polymersomes.Polymersomes were loaded contents through electroporation after polymersomes formed.The results showed that contents out of polymersomes and concentration reduced after reverse electroporation.The contents loaded efficiency of electroporation was much higher than traditional method.(3)The influence factors research of releasing and loading contents by electroporation.Loading contents by electroporation influenced by polymersomes properties,drugs properties and electrical pulse parameters.Electroporation numbers,voltage,hydrophilic and hydrophobic properties and molecular weight of contents,polymersomes concentrations were changed respectively.The results showed that voltage and molecular weight of contents had little influence in drug release and delivery.Electroporation numbers,contents properties,and polymersomes concentrations had huge influence in drug release and delivery.
