Oviductus Ranae is a traditional nourishing product in China.It contains many components such as protein,lipids,vitamins,minerals,and hormones.It has rich nutritional value and remarkable health functions.It has a reputation of “green soft gold”.Therefore,the research on Oviductus Ranae has continued in recent years,mainly including the composition and function of Oviductus Ranae.Protein as a major component of Rana sylvatica oil is a hot topic of research.However,the research on the activity of Rana sylvatica oil is obviously inadequate,and the Rana sylvatica oil product developed is even more limited.How to develop and use Rana sylvatica oil scientifically and reasonably still needs a lot of basic research to provide support.In this paper,through literature reading and experimental studies,the hydrolysis conditions of Rana oleifera protein were determined,and on this basis,the antioxidant activity,antitumor activity and antibacterial activity of Rana sylvatica oil peptides were studied.The specific findings are as follows:(1)Among the three kinds of single enzymes and two kinds of complex enzymes,the combination of papain and neutral protease obtained the best antioxidant activity of Rana oleracea L.,followed by single factor experiment and orthogonal optimization.The experiment finally determined that the enzymatic hydrolysis conditions of the composite protease were: the enzyme amount was 3%,the hydrolysis time was 7 h,the hydrolysis temperature was 55 ° C,and the hydrolysis pH was 7.0.(2)Polypeptides prepared from papain and alkaline protease were separated by ultrafiltration and separated into four molecular weight peptides >10 kDa,3-10 kDa,1-3 kDa,and <1 kDa,among which 3-10 kDa.The ability of Rana sylvestris oil polypeptides to scavenge free radicals was significantly higher than that of the other three peptides.When the peptide concentration was 10 mg/mL,the scavenging rates of hydroxyl radical,superoxide anion,and DPPH radical were 43.15±1.68% 、41.03±1.30% 、 58.06±1.71%.Compared with non-ultrafiltration isolated Rana oil mixed peptide,its antioxidant capacity has been respectively improved(p<0.05).(3)After different concentrations of Oviductus Ranae peptides interfered with the oxidative damage of L02 hepatocytes,the cell survival rate increased with the increase of the concentration of Oviductus Ranae peptides.When the polypeptide concentration was up to 4 mg/mL,the cell survival rate was 85.02± 4.27%,the survival rate of the model group was significantly higher(p<0.01)in the detection of glutathione peroxidase(GSH-PX)and catalase(CAT)activity,In both the medium and high dose groups treated,the activities of both enzymes were significantly increased(p<0.05).(4)The different molecular weight peptides prepared in the experiment did not produce inhibitory zone against Salmonella,Staphylococcus aureus,Escherichia coli and Listeria monocytogenes;in the minimum inhibitory concentration experiment,the experimental group containing the peptide showed turbidity.Phenomenon,these results indicate that the Oviductus Ranae peptides has no inhibitory effect on the above four bacteria.(5)The Oviductus Ranae peptides prepared in the experiment had no significant effect on the growth and proliferation of HepG2 and MCF-7 cells,but inhibited the proliferation of Caco-2 cells.When the concentration of polypeptide was 4 mg/mL,the inhibition rate was 18.35±1.21%%.
