Dietary fiber is not digested and absorbed by the body and similar edible carbohydrate foods,including hemicellulose,cellulose,pectin,lignin,gums,known as the following fat,carbohydrates,protein,minerals,vitamins and water after the "seventh largest nutrients".It has to obesity,high cholesterol,atherosclerosis,diabetes,colon disease and cardiovascular disease,certain preventive effect,improve the immune system and other physiological functions there.Where in the lignin also has a variety of biological activity,such as anti-oxidation,anti-tumor,anti-viral,regulate lipid metabolism,prevention of cardiovascular diseaseLignin is a phenylpropane unit by a carbon-carbon bonds of complex three-dimensional network?CC?and an ether bond?-0-?connection,amorphous phenolic polymer.It is one of the main ingredient of dietary fiber.Grape pomace?GP?was generated in the winemaking process to the grape skins,stems and grain as the main component of waste,accounting for about 20%of the grapes processing quality.Therefore,this paper grape pomace structural nature and effect of raw material,lignin dietary fiber properties,providing an initial experimental basis for the comprehensive utilization of grape pomace high value,while the prevention of many human diseases has far-reaching significance.As the following result::?1?The Polyphenols of wet residueGrape was 6.018mg/g,dry goods content of 8.858mg/g,its extraction process?decoloration?:at 90? condition,with 50%ethanol,solid-liquid ratio of 1.7 or wet residue grape skin extract three times,each time extracting 2.5h,extraction rate was 83.85%;grape pomace constituents decolorized:moisture 4.15%,23.43%protein,1.45%fat,5.69%ash,7.18%starch.?2?Decolorized grape pomace dietary fiber?the TDF?preparation:? The protein removal:liquid ratio 1:13 by adding distilled water,pH was adjusted to 10 by adding 1.0%protease solution,60? extracted two hours,washed with water to remove acid;? cooled to 50?,according to solid-liquid ratio 1:11,1%cellulase,hydrolysis 1.5h,hot water extraction 1.5h,heating to inactivate the enzyme,cooled to room temperature,add 4 volumes of ethanol and 100?,centrifuged,dried and weighed,extraction rate was 75.28%? Preparation of soluble dietary fiber:enzymatic product was placed in 100? water bath for one hour after the extraction,hot filtered and the filtrate concentrated to 1/3 of its original volume,cooling to room temperature.Adding ethanol filtrate was adjusted to a degree of 85%alcohol,allowed to stand at room temperature,filtered and dried,to SDF,extraction rate was 18.80%.Filter residue was dried,to the IDF,and weighed.Preparation,extraction rate was 41.36%?lignin:solid-liquid ratio of 1:30 was added 78%sulfuric acid solution,20? or 20? water bath shaker 2h,added 10 volumes of distilled water,heated to boiling cooker 1h,cooled,filtered,heated,dried and weighed,extraction rate was 27.71%?3?Determination the Property of TDF,IDF,TDF and LCC,its water holding capacity?WHC?was 5.354 g/g,5.162g/g,7.027 g/g,3.372g/g;expansion force?SWC?was 4.993 g/mL,5.196g/mL,3.972 g/mL,2.725g/mL;binding power?WBC?was 2.600 g/g,5.427 g/g,5.283g/g,11.759g/g;oil holding force?the OHC?,respectively to 4,429 g/g,7.459 g/g,4.494g/g,3.405g/g;cation adsorption capacity was 1.73 mmol/L,0.56 mmol/L,0.800 mmol/L,0.25mmol/L;in the small intestine,cholesterol adsorption capacity was 27.73 mg/g,26.40 mg/g,20.73mg/g,11.33mg/g,Cu2 + adsorption capacity was 2.460 mg/100g,23.520mg/100g,22.25 mg/100g,25.278mg/100g,Cd2 + adsorption capacity were 1.200mg/100g,6.990mg/100g,9.540mg/100g,5.161mg/100g,in the stomach,cholesterol adsorption capacity were 15.67 mg/g,8.17 mg/g,17.00mg/g,8.33 mg/g,Cu2+ adsorption capacity were 3.970 mg/100g0.810 mg/100g,3.180mg/100g,Cd2 +adsorption capacity of 21.260 mg/100g,13.890 mg/100g,17.170mg/100g,5.161mg/100g.SDF at 100 ? solubility of 88.46%;hydroxyl radical scavenging capacity of 90.38%.?4?SDF,IDF,TDF structure prepared by hydrolysis and acid hydrolysis lignin preparation have changed,the determination on the nature of the comparison results of lignin is part of the dietary fiber,affecting Property dietary fiber,dietary fiber definition further explainsnear the characteristic absorption peak of the benzene ring,stored in the TDF,IDF and lignin.whereas SDF does not exist,but cellulose-containing glucose,glycogen C-O bond.
