| Natural enzymes have attracted much attention because of their high catalytic efficiency and strong response characteristics.However,natural enzymes have problems such as inactivation and denaturation,difficulty in purification,high cost and difficulty in storage.Cancer is a serious threat to human life and health.Traditional cancer therapies are more or less damaging to human health.The key to improving patient survival is to use new drugs with higher anticancer activity for treatment.The metal organic framework(MOFs)has the advantages of large specific surface area and diverse skeleton structure.It has wide application prospect in energy storage,small molecule separation,drug loading and sustained release and heterogeneous catalysis,and it has great potential in the field of biology and materials.Therefore,it is very challenging to develop MOFs with mimic enzyme activity and anticancer activity.In the thesis,MOFs were used as materials to study the peroxidase and protease and to carry out simple anti-cervical cancer cell activity.The aim of this study was to explore the MOFs with high mimic enzyme activity and anti-cervical cancer cell activity.The simulated enzyme activity of 5%Fe/Fe-MIL-101 and FJU-21 was studied by enzyme kinetic method.The specific contents are as follows:(1)Fe-MIL-101 was synthesized by hydrothermal method,then Fe/Fe-MIL-101 with different loading was prepared.The crystal structure of 5%Fe/Fe-MIL-101 was characterized by scanning electron microscopy(SEM),X-ray diffraction(XRD)and specific surface area(BET).The peroxidase activity of 5%Fe/Fe-MIL-101 was studied by kinetic study and compared with Fe-MIL-101.It was found that 5%Fe/Fe-MIL-101 was more effective than Fe-MIL-101 has a strong affinity for ABTS and H2O2 under pH 4.0,50β conditions,and the Km value(0.095 mM)of 5%Fe/Fe-MIL-101 to ABTS is 9.6 times lower than the Km value(0.916 mM)of Fe-MIL-101.(2)FJU-21 was synthesized by solvothermal method.The crystal structure of FJU-21 was analyzed by XRD and BET.The effects of reaction pH,temperature and reaction time on the catalytic activity of FJU-21 were studied by simulating protease catalyzed hydrolysis of bovine serum albumin.The steady-state kinetics analysis of FJU-21 mimetic protease showed that its Kcat/Km(9.69 s-1M-1)was slightly higher than Cu-MOF Kcat/Km(7.73 s-1M-1).The Kcat/Km(3.72 s-1M-1)of FJU-21 was slightly higher than Cu-MOF(2.23 s-1M-1)at 50β.The catalytic activity of FJU-21 at 70β and 50βis better than Cu-MOF.FJU-21 can be recycled 11 times,indicating that FJU-21 is relatively stable.(3)The cytotoxicity of Fe/Fe-MIL-101 and FJU-21 on cervical cancer(HeLa)was determined by IC50.It was found that the inhibitory effect of Fe/Fe-MIL-101 and FJU-21 on HeLa cells were stronger than oxaliplatin(OXA)at 24 h,but the inhibitory effect were weaker than OXA at 48 h and 72 h.The results of 24 h,48 h and 72 h showed that Fe/Fe-MIL-101 and FJU-21 did not yet have good anti-HeLa cell activity. |
