Preparation And Medical Application Of Calcium Hydroxide Sustained-release Enteric Capsules

The common antipyretic analgesic and anti-inflammatory drugs mainly include non-steroidal anti-inflammatory drugs,steroidal anti-inflammatory drugs and heat-clearing-detoxifying herbs.Non-steroidal anti-inflammatory drugs can damage gastrointestinal tract,liver,kidney,cardiovascular and central nervous system.If we take the steroidal anti-inflammatory drugs for a long time,we would suffer from bone density loss,muscle atrophy,metabolic disorders and other adverse reactions.The composition of heat-clearing-detoxifying herbs is complex and the content of active ingredients is low,which make the efficacy of drug very slow.Meanwhile,the resources of heat-clearing-detoxifying herbs are found progressively decreasing in our country.In addition,the increasing number of patients with chronic disease affects people’s daily lives and health seriously in today’s society.Most of the chronic diseases associated with inflammation.Based on the above situation,this paper investigates a new type of alkaline anti-inflammatory drugs(calcium hydroxide sustained-release enteric capsules)which had an effective potency,and combined the health care and treatment efficacy.In the preparation of calcium hydroxide sustained-release enteric capsules,the loading capacity of calcium hydroxide should not be higher than 5.5 mmol g-1 for medication safety.Activation index and moisture content should be kept in range of 45%～50%and 5%～8%,respectively.And its solubility was 10 mg·(100mL)-1.The half-life period declined with the increase of loading capacity.There were three stages of the sustained-release processes,which all accorded with zero order release model.Acute toxicity test was carried out on rabbits in order to identify whether the calcium hydroxide sustained-release enteric capsules had any side effects or not.Even if the dosage of calcium hydroxide sustained-release enteric capsules was up to 2000 mg·kg-1,the results still showed that none of toxic effects had been occurred in the rabbits.The toxicity of calcium hydroxide was reduced and affinity between montmorillonite and mucous membrane was increased by loading calcium hydroxide on acid montmorillonite and modifying its surface.So the calcium hydroxide sustained-release enteric capsules were safe and non-toxic.Calcium hydroxide sustained-release enteric capsules conducted excellent performances in fields of antipyretic and anti-inflammatory.In the antipyretic and anti-inflammatory experiments,different doses of calcium hydroxide sustained-release enteric capsules(12.5 mg·kg-1,25.0 mg·kg-1)could reduce the rabbits’ fever symptoms and the levels of inflammatory factors(IL-6,TNF-α).Its effect of antipyretic and anti-inflammatory was significantly superior to aspirin.Besides antipyretic anti inflammatory,calcium hydroxide sustained-release enteric capsules were very effective on promoting digest,anti-alcohol and anti-inebriation.The rabbit digestion experiments demonstrated that the digestibility of dry matter,protein,crude fat,crude ash and calcium apparently increased by 28.6%,13.3%,15.9%,30.9%and 24.2%,respectively.There was no increasing trend on the rabbit’s intake,which indicated that calcium hydroxide sustained-release enteric capsules were conducive to digestion and absorption of nutrients.According to the anti-alcohol and anti-inebriation experiments,calcium hydroxide sustained-release enteric capsules were very effective on anti-alcohol and anti-inebriation.The drunk sleeping time of anti-alcohol and anti-inebriation group was 163 min and 125 min,respectively,demonstrating a significant advantage over model group,whose sleeping time was 259 min.The capsules were helpful to restore the digestive function and accelerate the catabolism of alcohol in the body and reduce the injury which caused by alcohol and intermediate products to gastrointestinal tract,liver and other organs.It was indeed a kind of excellent health care products.Intensity and solubility of alkali were reduced by loading calcium hydroxide on montmorillonite.Calcium hydroxide was firstly encapsulated in enteric capsules to use in small intestine.The capsules performed excellent properties of antipyretic,anti-inflammatory,promoting digestion and absorption,anti-alcohol,anti-inebriation and so on.It was a new type of alkaline drugs to treat and prevent inflammatory.In addition,calcium hydroxide sustained-release enteric capsules also had certain application value in calcium supplements,relieving constipation,and reducing blood pressure.
