| In order to reduce the environmental pollution caused by livestock manure in the countryside and the environmental pollution caused by heavy metals in the livestock manure.In this study,a new microbial inoculum for livestock manure composting was selected,and then a bio-organic fertilizer suitable for plant growth was developed based on it.First,the screening of the desulfurization and deammoniation strains and passivating heavy metal strains of livestock manure,the identification of high efficient strains,and the optimization of the combination of deodorizing strains and heavy metal passivation strains,and developed the degrading bacteria of livestock manure and inoculated the degrading bacteria were carried out.According to the growth needs of sunflower,N,P,K and other elements were added to obtain the special bio-organic fertilize suitable for sunflower growth and its application effect on sunflower.The results are as follows:(1)By the analysis of the external morphology of the strains and the BLAST comparison analysis of the 16S r DNA fragment revealed that.Alcaligenes faecalis,Proteus mirabilis,Bacillus cereus,Bacillus siamensis,Streptomyces alboflavus,Nocardioides terrae.(2)The results showed that the growth of maize were inhibited under heavy metal chromium(Cr2O72-)stress(50 mg·kg-1 and 100 mg·kg-1),inoculation of Alcaligenes faecalis.qz-1 could significantly alleviate the adverse effects of heavy metal stress on Maizeg rowth.At the same time,inoculation with Alcaligenes faecalis.qz-1 could obviously increase the uptake of chromium(Cr2O72-)by maize,the total uptake increased by 13.9%-36.9%compared with non-inoculated plants.In summary,inoculation of Alcaligenes faecalis.qz-1 improved the growth characteristics of maize under the heavy metal chromium(Cr2O72-)stress,increased the uptake of chromium(Cr2O72-)in maize roots,and reduced the toxicity of chromium(Cr2O72-)to Maize,reduced the pollution of chromium(Cr2O72-)to soil.(3)In this study,we identified the strains with strong ability to degrade biological macromolecules screened by our research group,obtained 4 strains of bacteria,2 actinomycetes with high degradation activity,fast growth rate,optimized and combined,and developed 2 groups of degrading strains of livestock manure.The 2 livestock manure degrading strains were added into the heap,and their water content,temperature,p H,and organic matter content were measured.Compared with the control group,the bacteria with good degradation effect were obtained.The results showed that the composting time of decomposing bacteria was better than that of control group.Therefore,inoculation of livestock manure degrading strains can promote composting.(4)A pot experiment was conducted to study the effects of bi-organic fertilizer and amount of sunflower on plant height,stem diameter,chlorophyll content,aerial part,underground dry weight of sunflower.Under the condition of sunflower bio-organic fertilizer(0.40g/kg,0.50g/kg)and compound fertilizer application(0.50g/kg),the growth of sunflower was obviously better than that of the control(p<0.05),for the plant height increased by 29.15%,15.39%and 19.43%,the stem diameter increased by 31.25%,8.33%and 25%,the chlorophyll content increased by 19.8%,7.20%and 12.9%,the root dry weight increased by 27.57%,19.6%and 23.92%,the stem weight increased by10.31%,7.37%and 8.41%,the dry weight of leaves increased by 14.71%,4.90%and5.88%respectively.The experimental results show that the effect of bio-organic fertilizer is significant,The livestock manure bio-organic fertilizer developed in this study can be applied to the plant by adding a proper amount of nutrient elements in the bio-organic fertilizer,which provides a theoretical basis for the plant fertilization according to the demand for various nutrient elements for different plants. |
