Paeonia ostii t.Hong et J.X.Zhang is as a category of paeonia suffruticosa in Tongling.In 2011,paeonia ostii and paeonia rockii seed oil were approved as a new resource food by the Ministry of Health.The content of unsaturated fatty acids in peony seed oil is rich,especially the alpha-linolenic acid and linoleic acid.The liposoluble constituent have some influence on the nutritional quality and function of edible oil.The composition and composition of unsaturated fatty acids are also significant for tumor cells.In this paper,the content of total phenol,squalene,vitamin E,β-glutosterol and polyphenol in paeonia ostii seed oil was determined by folin-ciocalteu method,GC method and HPLC method.The composition and content of fatty acids in paeonia ostii seed oil and olive oil were determined by GC/MS.The two were reconciled in a certain proportion.The CCK-8 method was used to determine the tumor cell inhibition rate as an evaluation index.The effect of the harmonization of paeonia ostii seed oil on HepG2 tumor cells was investigated in order to provide a theoretical basis for the deep use of paeonia ostii seed oil.The main results were as follows:1.Analysis and determination of the liposoluble constituent of the paeonia ostii seed oil The content of total phenol in paeonia ostii seed oil was determined by folin-ciocalteu method was 16.58μg·g-1.The content of squalene in paeonia ostii seed oil by GC method was 55.70μg·g-1.Measured liposoluble constituent and content of paeonia ostii seed oil by the method of HPLC is respectively:α-tocopherol 409.10μg·g-1,γ-tocopherol 194.10μg·g-1,β-sitosterol1794.30μg·g-1,epicatechin1.58μg·g-1,catechins 1.92μg·g-1,chlorogenic acid 1.06μg·g-1,rhizoma kaempferiae 8.48μg·g-1,therefore,the liposoluble constituent in paeonia ostii seed oil are diverse,rich in content and high in nutritional value.2.Analysis of the composition of paeonia ostii seed oil and olive oil fatty acid(FA).The content of unsaturated fatty acid(UFA)of paeonia ostii seed oil and olive oil was relatively high,85.81%and 78.09%respectively.The contents of paeonia ostii seed oil accounted for 65.51%ofα-linolenic acid,18.65%of linoleic acid,9.07%of palmitic acid,and 2.48%of stearic acid.It can be seen that the unsaturated fatty acids in paeonia ostii oil are mainly unsaturated fatty acids(PUFA);The olive oil is 71.56%of oleic acid,16.27%of palmitic acid,5.37%of linoleic acid,and 4.62%of stearic acid.Therefore,unsaturated fatty acids in olive oil are mainly monounsaturated fatty acids(MUFA).The content ofα-linolenic acid and linoleic acid in paeonia ostii seed oil was determined by GC method,respectively,539.50 mg·g-1 and 296.70 mg·g-1.3.The inhibitory effect of paeonia ostii seed blend oil on HepG2 in vitro tumor cells.(1)In the experiment,different doses of paeonia ostii seed oil and olive oil had inhibitory effect on HepG2 in vitro tumor cells,and with the increase of dose concentration,the dose-effect relationship was obvious.The IC50 of paeonia ostii seed oil is 6.05μL·mL-1,and the IC50 of olive oil is 6.01μL·mL-1.The inhibition of HepG2 in vitro tumor cells was similar.(2)According to the analysis results of the composition of paeonia ostii seed oil and olive oil,they were made into a blend of different MUFA and PUFA ratio,investigation of its influence on HepG2 in vitro tumor cell inhibition rate found that MUFA with different ratio of PUFA has inhibitory effect on HepG2,compared with a single oil,blend oil inhibition rate increases,and the inhibition rate order from high to low proportion of 1:1,2:1,1:2,3:1,1:3.
