With the problems of food pollution increasingly became serious.animal origin-food safety is put highly sight on people and become one of universal problems faced all over the world.In order to comprehend the condition of pollution in immigration control of people who carry animal origin-food.Eight kinds of animal origin-food in Helgas port are tested by the national standard,which contain sausage,canned meat,milk beverage,edible animal oil,honey,canned or nitrite,coliform,Salmonella and STEC.Result shows that five that five Salmonella are tested in 66 random inspections where one is in canned meat,one in milk cream and another on in honey and the rest two are tested in edible animal oil showing that relevance ratio is up to 7.5%.Salmonella pollution are serious in animal origin-food carried by port entering passengers in Helgas port.Salmonella,LM and STEC O157 are three main food-borne bacteria in meat and meat products.This research develops Salmonella,LM,STEC O157 these three kinds of bacteria to test.We choose specificity genes individually:invA,hlyA and rfbE as target genes of three target bacteria,we optimize primers ratio and annealing temperature of sensibility of reaction system.Finally,Screening of the added amount of optimal primer,and appropriate annealing temperature is 58?.The sensibility is 10 CFU/mL,10 CFU/mL,10~2 CFU/mL,three kinds of pathogenic bacteria is 10~3 CFU/mL,In this reaction system,we find that Sa,CJ and Shigella have no significant amplified bands,indicating strong specificity.And we collect287 samples of mutton,beaf and Trunks swab in market and shambles in Kuerle,Using treble PCR and traditional method test three kinds of pathogenic bacteria.We find that 9 Salmonella,positive rate 3.2%and 2 LM,relevance ratio is 0.7%and 2 STEC O157,relevance rate is0.7%.Compared with the traditional bacterial identification method,the coincidence rate of the three pathogens was 96%,97.8%,99%.Optimized treble PCR in this paper can test Salmonella,LM and STEC O157 at the same time,reaching the target of rapid detection of Food-borne bacteria.
