Isochrysis galbana is a kind of marine microalgae,which is rich in fucoxanthin and unsaturated fatty acid.Fucoxanthin is an oxygen-containing carotenoid,it has the physiological activities such as antioxidation,regulation of blood glucose and anti-angiogenesis,and plays an important role in human health,beauty,food and so on.Unsaturated fatty acid has the function of regulating cardiovascular disease and brain function,and plays an important part in the process of human growth and development.This study provides some valuable references for subsequent industrialization production and research.The main contents and results of this paper were described as follows:1.Through the isolation and purification methods and antibiotic sterilization studies,we obtained the uncontaminated Isochrysis galbana.By means of different preservation methods,the preservation of Isochrysis galbana was studied,and the preservation method of liquid secondary preservation and solid plate test tube was better than that in Isochrysis galbana.By morphological and molecular biological identification,it was identified as a Isochrysis galbana in Isochrysis sp.;The genome size of Isochrysis galbana was estimated to be about 102.671 Mb by the BD FACSymphony A5.2.The process optimization of Isochrysis galbana fucoxanthin by organic solvent extraction,and obtained the best extraction process optimization combination for for extracting temperature of 35℃,liquid ratio of 1:30 g/mL,extraction time for 15 min,extraction times for 2 times,extraction yield of 12.03 mg/g;The best recovery rate was 86.35%and the purity was 29.78%,which the crude fucoxanthin extract of the Isochrysis galbana is separated and purified by silica gel column.Through UPLC and HPLC,which a method for the detection of fucoxanthin was established,the test results showed that the content of fucoxanthin in Isochrysis galbana was about 659 by LC-MS.3.Through the optimization of the cultivation conditions of Isochrysis galbana,it is concluded that the optimal temperature is 25℃,the light intensity is 120 μmol photons m-2 s-1,cycle for 14 h:10 h,salinity at around 31.5‰,PH value around 8.0,it is beneficial to the growth of Isochrysis galbana;The optimization of the culture mediums of Isochrysis galbana,the content of fucoxanthin increased by 2.54 times and the total oil content increased by 2.77 times.The production of fucoxanthin and lipids was increased by increasing the aeration of 5 L triangulation bottles and 50 L photobioreactors;The best flocculant of ferric chloride through the flocculation and harvesting of Isochrysis galbana,and it was beneficial to the recovery after harvesting.4.The results showed that there was no significant influence on the accumulation of fucoxanthin and fatty acid metabolism in Isochrysis galbana by different light quality and nitrogen and phosphorus stress.Seven fast growth and high fucoxanthin and high oil productivity mutants of Isochrysis galbana were screened by UV and ARTP mutation breeding technology.
