Enzyme is a kind of catalytic material produced by the organism,because of its high efficiency,specificity and gentleness,it has attracted more and more attention and has been applied in many fields,such as food industry,washing industry,pharmaceutical industry and so on.With the further development of science and technology,enzyme will be more widely used.However,there is a problem here.Although the enzyme can be isolated from the organism,there are many problems,such as cumbersome cultivation and subsequent purification methods,complex separation process,many technological procedures and large energy consumption,which all restrict the application of enzyme preparation.In this paper,the microbial fermentation of alkaline protease 2709 was studied.Bacillus lichoniae 2709 was used for fermentation culture.On the basic fermentation medium,a better fermentation medium formula was selected by changing different carbon sources,nitrogen sources and inorganic salts,and the basic medium was improved.At the same time,bacillus lichydiae 2709 was fermented with improved medium to obtain the fermentation broth of 2709 alkaline protease.For the 2709 alkaline protease fermentation broth,because the improved fermentation medium is composed of corn powder and soybean powder,the final fermentation broth is thick and contains many impurities.Essentially,2709 alkaline protease zymotic fluid is a kind of "water",we choose to use polyacrylamide,aluminum sulfate and ferrous chloride solution,to deal with fermented liquid,and on the basis of single factor experiment,the optimum adding amount of three kinds of solutions,using the orthogonal experiment,obtain the optimal ratio of compound,namely polyacrylamide(400 w,0.5%),6 ml,aluminium sulfate(10.0%),0.3 ml,ferrous chloride(10.0%),1.2 ml,in proportion to the compound adding coagulant,and used in the processing of fermented liquid,finally get the processing of the liquid,The loss of enzyme activity was between30% and 40%,and the clarification was increased by about 70%.Concentrated 2709 alkaline protease solution was obtained by certain treatment after obtaining high clarification fermentation liquid.We added it into the soap to study the decontamination performance of 2709 alkaline protease in the soap,and obtained theoptimal addition amount of 2709 alkaline protease in the soap,and explored the stability of 2709 alkaline protease in the soap.When enzyme stabilizer was not added to the soap group,the activity of alkaline protease 2709 remained about 10% after 7 days,while the retention rate of enzyme activity was still 30% within 60 days after the addition of enzyme stabilizer.
