In this paper,the traditional Chinese medicine material Mgnolia liliflora was used as raw material,and the extraction process of microwave assisted extraction stage was carried out by using the response surface method by using microwave-assisted extraction method to extract pinoresinol dimethyl ether,fargesin and magnolin from Magnolia liliflora.Optimization,the use of macroporous resin for the enrichment and purification of pinoresinol dimethyl ether,caprylic and magnolin in Magnolia liliflora.Its main contents are as follows:(1)A high performance liquid chromatography(HPLC)method for the determination of pinoresinol dimethyl ether,fargesin and magnolin was established.The chromatographic conditions were:detection wavelength:283 nm;mobile phase composition and ratio:acetonitrile:water=43:57;column:Kromasil C18(5 μm,4.6 mm x 250 mm),flow rate:1.0 mL/min,injection volume 10 μL,column temperature:28℃,isocratic elution for 30 min.The pinoresinol dimethyl ether,fargesin and magnolin have a good linear relationship within the range of the measurement.(2)The method for extracting pinoresinol dimethyl ether,fargesin and magnolin was determined.The response surface optimization method was used to optimize the extraction conditions on the basis of single factor test computer.The different solvent types,extraction concentration,solid-liquid ratio,homogenization time,microwave pretreatment time and power were compared.The optimal extraction conditions were determined:the optimal extraction conditions were:extraction solvent 1,4-butanediol,solvent concentration 40.02%,liquid to material ratio 11.84:1,microwave power 230 W,microwave time 24.97 min.The total yield of the three lignans was 183.05 mg/g.(3)By comparing the adsorption and analytical properties of the eight kinds of macroporous resin,it was confirmed that the adsorption and resolution of the resin lignin D101 on the three kinds of lignans were higher than those of other resins.The optimum adsorption parameters of resin D101 were determined:the optimum sample loading was 200 mL,the adsorption temperature was 25℃,the loading flow rate was 2 BV/h,and the amount of resin adsorbed saturated pinoresinol dimethyl ether was 14.56 mg/g.The amount of the fargesin was 12.22 mg/g,and the amount of the magnolin was 80.06 mg/g.Pine determined the optimal desorption series parameters of macroporous resin D101:70%ethanol solution is a dynamic elution desorbent.The desorption rate of rosin dimethyl ether can reach 88%,the resolution of capsin can reach 85%,and the resolution of magnolin can reach 90%(4)Microwave-assisted extraction of rosin dimethyl ether from Magnolia liliflora,which is a high-efficiency,rapid,non-polluting and high-yield extraction method.This experiment confirmed its ability in the field of natural medicinal chemistry.Developmental,providing a more feasible solution for the extraction of target products.
