Purpose Donkey-hide Gelatin,included in the Chinese Pharmacopoeia(Page 189 of the 2015 edition),has been used as a medicine 2500 years.“Gelatin is both food and medicine,which is the same as medicine and food”,the ministry of health stipulates in the Notice on Further Regulating the Management of Hearth Food Raw Materials published in February 18,2002.Donkey-hide glue is not only used as a traditional Chinese medicine in the treatment of clinical diseases,but also made into a variety of health food to enhance human immune function.At present,the gelatin health foodin markets includes products as follows:tablet,powder,walnut cake,paste,oral liquid,capsule,jujube,lozenge and so on.With the rapid increase in the use of the raw materials and the lack of medicinal resources,the price of Donkey-hide gelatin has been increasing in recent years,which has led to gelatin adulteration,for instance,some manufacturers use cowhide,pigskin or even the raw material of the industrial revolution to replace donkey hide.The the Chinese Pharmacopoeia(2015 edition)provides a method for identifying gelatin and its content determination.The No.2012001 Approval of CFDA Supplementary Inspection Methods and Items for Drug Inspectionspecifies the supplementary test method for the source of cow-hide skin in Donkey-hide gelatin.There is no special quality standard for Donkey-hide gelatin products as health food,so it is necessary to establish a method for its quality evaluation and control.The purpose of this research is to test samples in markets and evaluate the quality of donkey-derivedhealth foods,by detecting the situation of adulteration and harmful heavy metal content.Method In this research,UPLC-MS/MS method was established to detect ingredients of donkey-hideand cow-hide source in markets’ gelatinhealth foods,6 kinds of dosage forms of 61 batches;In the sample preparation for 7 kinds of dosage forms of 100 batcheswas carried out by microwave digestion.The content of arsenic and mercury was determined by atomic fluorescence spectrometry and the content of lead and chromium by atomic absorption spectrophotometry;Pig-skin gelatin is the larger proportion in the adulterated gelatin,and there is no identification method of this new Gelatin.Under that circumstance,high resolution mass spectrometry was performed after enzymatic hydrolysis of donkey skin and pig skin with the help of protease digestion and mass spectrometric peptide recognition technology.Results There is nothing but the characteristic peak of gelatin component was found in 22 batches of samples,only the characteristic peak of cowhide source component,25 batches.Both characteristic peaks concerned above were simultaneously detected in 8 batches of samples,Neither one was found in 6 batches.The results illustrate that this method can quickly identify whether the sample is a health food with Donkey-hide ingredients,and whether it contains cow-hide sources,which can greatly accelerate the inspection efficiency of donkey-hide health food;At the same time,this method can also quickly find the phenomenon of "genuine products mixing with fake products";The results show that the chromium content meets national standards,and the contents of three heavy metals including mercury,arsenic and lead in oral liquids and pastes were higher,which might be due to the high residual content of mercury,arsenic and lead in the added Chinese patent medicine;The coverage rates ofmm the corresponding species were 71.0% and 49.5% respectively,by comparing the collected high-resolution peptide fingerprint data and the established collagen database.Conclusion In this research,UPLC-MS/MS method was established to detect ingredients of donkey-hide and cow-hide source in markets’ gelatinhealth foods;The content of arsenic and mercury was determined by atomic fluorescence spectrometry and the content of lead and chromium by atomic absorption spectrophotometry;Protease digestion technique and mass spectrometry polypeptide recognition technology help to seek characteristic peptide sections of pig-skin gelatin which is different from donkey-hide gelatin.That can be used for the identification of the new adulterated gelatin and offer the research thought and experimental evidence for quality standard establishment of Donkey-hide Gelatin Healthy Food.
