| Tobacco,as an important economic crop in the world,is used in the cigarette industry and various components extracted by it,have also applied in the fields of medicine and pesticides.Headspace Solid Phase Microextraction(HS-SPME)is an extremely simple and efficient enrichment technology for detecting the volatile and semi-volatile components,it saves the volume of solvent and shortens the pretreatment time during operation.In additon,the enrichment factor of components can be up to 2-3 orders of magnitude.Enhanced capacities of qualitativeandquantitativeanalysiscapabilitiesofGas Chromatography-Mass Spectrometry(GC-MS)make it the first choice for the separation and identification of the volatile and semi-volatile complex mixtures.Based on the high enrichment ability of HS-SPME and the powerful qualitative as well as quantitative capability of GC-MS,in this paper,we studied the aroma characteristics of tobacco,including the possibility of fragrant discrimination and cigarette flavor analysis.Main works were showed as follows:1.The extraction and enrichment ability of tobacco volatile and semi-volatile components were compared by HS-SPME and conventional steam distillation(WSD)method.The composition of tobacco samples was obtained by GC-MS.The results showed that the HS-SPME method can get more information of tobacco samples.In addition,the extraction conditions were optimized,including the extraction head material,the amount of samples,the extraction time and extraction temperature by HS-SPME method,and the repeatability was investigated.2.Under the optimized conditions,scent typing on 140 batches of flue-cured tobacco samples was studied by HS-SPME-GC-MS combined with the chemometrics software ChemPatternTM,principal component analysis(PCA)and cluster analysis(CA)were used to data processing of various flue-cured tobacco produced in different areas.The results show that the fresh-sweetness aroma,the honey-sweetness aroma and the other scent types can be distinguished clearly,and there was a difference in aroma characteristics between the flue-cured tobaccos of different areas.The results also showed that HS-SPME-GC-MS combined with metrology method can effectively distinguished different flavors of flue-cured tobacco.3.HS-SPME-GC-MS technology combined with the chemometrics software ChemPatternTM was used to analyze the aroma components in mixed and flue-cured cigarettes.CA and PCA images were obtained,while there were two obvious categories exists in the CA image,the PC1and PC2 scores were 53.30%and 40.02%in the PCA image,respectively,which can explain almost all the original data.Several significant components that contributed a lot to the classification were explored,which means the types of cigarettes can be distinguished effectively. |
