With the rapid development of economy,drug abuse is becoming more and more serious,which has a great impact on the drug use of patients and social security.Especially with the emergence of a series of new synthetic drugs,the national drug monitoring and testing are facing enormous challenges.At present,most of the detection methods used in our country have the disadvantages of large volume,high cost and complex pretreatment.Surface Enhanced Raman Spectroscopy(SERS)has been widely used in trace analysis,biological identification,food safety and other fields due to its high accuracy and short detection time.Silver sol was prepared as the active substrate,and its morphology and properties were characterized.On this basis,three psychotropic narcotic drugs,fentanyl citrate,sufentanil citrate and midazolam,were tested.The optimized conditions were explored,which laid a good foundation for the supervision of special drugs.Firstly,the molecular structure of fentanyl citrate,sufentanil citrate and midazolam psychotropic narcotic drugs was optimized by using the software Gauss 09.The characteristic peaks of the three drugs were assigned based on the density functional theory,and the measured and calculated spectra were compared to determine the characteristic peaks of the three drugs.Then,SERS substrates with good stability and high sensitivity were prepared by Lee and Meisel methods.The properties of the silver particles were characterized.Most of the silver particles obtained were 40-50 nm in size,spherical in shape and uniformly distributed.The results show that the silver sol has good SERS enhancement effect.Finally,water and urine samples of fentanyl citrate,sufentanil citrate and midazolam were prepared.The silver sol was used to optimize the detection conditions of the three drugs.At the same time,the repeatability of the experiment was validated by collecting the sample spectra under the optimum conditions.By analyzing the Raman spectra of three drugs with different concentrations,the fitting curve and detection limit of aqueous solution were obtained.The recoveries and relative standard deviations of samples with high,medium and bottom concentrations were calculated respectively.The curves with high recovery and good fitting effect show that this method is efficient and accurate,and will be widely used in the field rapid detection of narcotic drugs.. |
