Prevalence of cardiovascular disease increased progressively in the past few years,is closely related to the dietary lipids intake.Ingested dietary lipids,such as cholesterol,are absorbed and metabolized in the intestine with the form of ―Dietary Mixed micelle,DMM‖.In this article,lactone tofu and hard tofu were chosen as the desired test sample material to be digested,and the vitro digestive enzyme as the simulation mode,the samples were separated and purified by centrifugation and filtration.Detected by both Kit and HPLC-ELSD,basing on the influence of products‘ cholesterol micelle on transportation,the two matrix,namely artificial and natural pig bile micelles were explored to determine their effects of final detection results.Experimental data shows that tofu enzymatic digestion product via vitro digestion can inhibit the transportation of cholesterol micelle,and the capacity of inhibiting cholesterol‘s transport depends on the molecular weight of products.Comparing the different molecular weight product from lactone tofu and hard tofu,the micelle cholesterol absorption rate of >10k HHT was(55.34±0.03)%,manifesting the highest ability to inhibit the transportation of cholesterol micelles.3k ~ 10 k HHT and HLT played approximate role in the inhibition of cholesterol micelle transportation,whose absorptivity were respectively(75.35±0.14)% and(73.82±0.24)%.The results of Kit and HPLC-ELSD have consistency,which prove that both of them can be used to validate the activity of samples.Swine bile micelles can replace the manual preparation of micelles as cholesterol micelle experimental matrix,avoiding the time-wasting and the manual errors caused by during the preparation of the micelle solution.
