Resveratrol is a plant polyphenol extracted from grapes,which has strong biological activity.Since the discovery of resveratrol in 1993,there are many reports on it,but the real application of resveratrol is rare.It plays an important role in anti-inflammatory,anti-canner,anti-oxidation,anti-aging and platelet aggregation.However,the disadvantage of light decomposition and fast metabolism in vivo,which not only limit its storage of resveratrol,but also fails to reach the lesion site after oral administration,resulting in waste of drugs.Therefore,it is of great significance to explore suitable dosage forms,improve the efficacy of resveratrol and make it play its medicinal value.1.Resveratrol was modified with chlorophytum-shaped PCL.Poly(ε-caprolactone)(PCL)with molecular weight 6000 was synthesized by using ring-opening polymerization with pentaerythritol.And then PCL was acidified by succinic anhydride(SA)to prepare the acidification product PCL-COOH which was used to obtain the final product by esterification with resveratrol.The synthesis of product was proved by a series of characterization;meanwhile the photostability and the rapid metabolism of resveratrol were greatly improved.2.Resveratrol was anchored onto the surface of polylactide nanofibers.Nano-fiber membrane has the characteristics of simulating extracellular matrix,but it has no activity.Therefore,in this paper,polylactide fiber membrane is used to play the role of matrix,loaded with active resveratrol,to prepare biologically active nano-fiber membrane.The loaded-resveratrol was prepared by grafted resveratrol onto blended fiber membrane of PCL-COOH/PLLA.The synthesis of loaded-drugs fiber was characterized by nuclear magnetic resonance(NMR);the good morphology was shown by electron microscopy;the mechanical strength of fiber reached 1MPa;and it shows good infiltration and non-toxicity,so it was hopeful to be applied in tissue engineering.3.resveratrol and xanthophenol(XAN)was synergistically transporting by coaxial nanofibers.Polycaprolactone-resveratrol polymer(SP-RES)and polyethylene oxide(PEO)were used as hydrophilic core structure,XAN and polylactic acid-glycolic acid copolymer(PLGA)as hydrophobic shell structure.Through the comprehensive analysis of core-shell fiber infrared,electron microscopy,tensile force,hydrophilicity,cell viability and in vitro release,the hydrophilic core-hydrophobic shell coaxial fiber membranes which can synergistically deliver drugs were obtained.The optimum proportion was: 10wt% resveratrol-polymer/PEO(50/50)core content and 10wt% XAN/PLGA(10/90)shell content. |
