| As a kind of perennial herb,Tianshan Saussurea involucrata Kar is a very precious Chinese medicine resource,which is known as the “King of Herbs” and “The Best of Medicine”.In recent years,the cell culture of Tianshan Saussurea involucrate Kar has been cultivated by biotechnology,and the relevant researches towards this indicates that the Saussurea involucrata Kar cell cultures are full of Caffeyl Quinic Acid(CQA),which has the bioactivity of reducing blood fat and anti-obesity,but the mechanism of action is not clear.In this study,the purified CQA substance extracted from the cell culture of Tianshan Saussurea involucrata Kar were used as the object.This research made preliminary study on the anti-obesity mechanism of CQA substances through activity experiment of digesting enzyme inhibitory test in vitro & obesity suppression experiment in feeding mice.This research got the following conclusion:(1)Extracted and purified CQA substances in Saussurea cell cultures,through water extraction,macroporous adsorption resin and purification of prepared liquid,finally obtained purified CQA substances,which accounted for 3.5±0.75% of dry weight.(2)In vitro inhibition experiments of a-amylase,a-glucosidase and porcine pancreatic lipase on CQA substances,the results showed a positive correlation trend with the increase of the final concentration of CQA substances in the samples,and the inhibition rate of digestive enzymes were gradually increased.When the final concentration of CQA substances reached 1.5 mg/ml,the inhibition rate of a-glucosidase and porcine pancreatic lipase could reach 95.38%,97.1%;when the concentration reached 2.25 mg/ml,the a-amylase inhibition rate reached 100%.(3)The study on anti-obesity effect of high-fat fed mice showed that CQA substances in Saussurea cell cultures can effectively reduce the body weight,Lee’s index,liver weight and epididymal fat coefficient of high-fat rats.It can reduce the levels of Total cholesterol(TC),Triglyceride(TG),Low density cholesterol(LDL-C),Malondialdehyde(MDA),and Lipase(LPS)in serum and liver homogenate,as well as increase the level of High density cholesterol(HDL-C),Superoxide dismutase(SOD),and Glutathione peroxidase(GSH-PX),which can regulate fat metabolism disorders and reduce the formation of lipid peroxide,improve the formation of anti-peroxide and reduce the anti-obesity effect of fat absorption and accumulation in mice fed with high fat diet.(4)The results of using real-time PCR to detect the expression levels of key enzyme genes in fat metabolism of high-fat mice by CQA showed that CQA substances in Saussurea cell cultures could reduce the expression level of Fatty Acid Synthase(FAS)and Hydroxymethylglutaryl coenzyme A reductase(HMG-COA),increase the expression of peroxide-expressing activator receptor-α(PPARα)and activate the expression of cholesterol 7α-hydroxylase(CYP7A1),thereby making an anti-obesity effect. |
