As an ancient medicinal plant,Salvia officinalis L.has important edible and medicinal value,and its main active ingredients are essential oils and phenolic substances.In this thesis,the extraction of its volatile components(essential oil)and the extraction and purification of its non-volatile components(phenolic substances)are studied.The main results as follows:(1)In this study,dual-cooled solvent-free microwave extraction has been used for the extraction of S.officinalis essential oil,and the process parameters are optimized by Box-Behnken(BBD)experimental design: humidity 71%,extraction time 35 min and irradiation power 650 W,respectively.Under the optimized conditions,the predicted yield of essential oil is 1.602%,which fairly agreed with the observed yield(1.592%).Compared with hydrodistillation(0.856%)and SFME(1.296%),the dual-cooled SFME efficiently reduced heat loss,and presents a higher extraction yield.The components of essential oils extracted by HD,SFME and dual-cooled SFME have been analyzed by GC-MS.A total of 37 components are identified in the essential oils obtained by HD,SFME and dual-cooled SFME,which representeds 89.28%,92.50% and93.59% of the total oil composition,respectively.It has also found that the essential oils extracted by SFME and dual-cooled SFME contained higher amounts of oxygenated monoterpenes(67.06% and 68.00%)and lower amounts of monoterpene and sesquiterpene hydrocarbons(10.36% and 10.13%,6.75% and 6.78%).Antimicrobial activity analysis shows that S.officinalis essential oils can significantly inhibit Gram-positive bacteria,especially Bacillus subtilis and Staphylococcus epidermidis.Compared with HD,the essential oils obtained by SFME and dual-cooled SFME have better antibacterial effects.(2)In this study,integrated ultrasonic extraction-macroporous resin purification method has been used to extract and purify the phenolic substances from S.officinalis,and the process parameters are optimized by single factor experiment: macroporous adsorption resin type:D101-1;amount of macroporous adsorption resin: 3.0 g(ratio of resin to plant material is 1:1);extraction temperature: 40 °C;extraction time:1.0 h;flow rate: 35 BV/h;eluent: 3 BV 70%ethanol.Under this condition,the yield of phenolic substances is 59.58 mg/g.The yield of phenolic substances and its proportion in the extract in this method are 1.2 and 4.8 times those in ultrasonic extraction method,respectively.The dual-cooled SFME not only improves the yield of essential oil,but also better retains the aroma of essential oil and antibacterial activity.Thus,dual-cooled SFME was an effective improvement to traditional method.Integrated ultrasonic extraction-macroporous resin purification method is easy to operate,not only improves the yield of phenolic substances and its proportion in extracts,but also shortens the extraction time and reduces the use of organic reagents.It is a fast,green,and effective method for extraction and purification of phenolic substances from S.officinalis.This thesis provides scientific basis and technical support for the development and utilization of S.officinalis,and will further promote its application and development in the fields of medicine,food and daily necessities.
