In order to construct an ordered multi-enzyme catalysis system and realize efficient multi-enzyme co-catalysis.Inspired by the complement ary pairing mechanism of the nucleotide sequence,two multifunctional star polymers were designed and prepared by RAFT polymerization.The ordered multienzyme systems have been construted using multiple positioning characteristics of materials.This strategy provides a novel idea for ordered enzyme immobilization.Based on the functional monomers of N-adamantylacrylamide and N-succinimidyl acrylate,a functional star polymer was prepared using an“arm first”approach via RAFT polymerization.By adjusting the amount of crosslinking agent,the size and polydispersity of the star polymer were controlled.With the star polymer as the scaffold,a sequential multi-enzyme system was constructed,where horseradish peroxidase was placed in the inner layer and ?-cyclodextrin-modified glucose oxidase was assembled in the outer layer.In addition to the enhanced thermal stability,this multienzyme system also exhibited a reduced Km value(from 2.18 mM to 0.39 mM)and an excellent substrate affinity.More interestingly,the activity of the enzyme modified by b-cyclodextrin was improved.A block copolymer brush based on these functional monomers successfully grew on the surface of magnetic particles by surface-initiated RAFT polymerization.Similarly,horseradish peroxidase and glucose oxidase were constructed into a multienzyme system.After being reused 8 times,the activity of multienzyme system retained 67.21%of the initial catalytic activity.
