In recent years,dissolving microneedles have attracted extensive attention in the direction of transdermal drug delivery because of their advantages such as painlessness and convenience.Usually,because the elasticity of the skin will sag when the dissolving microneedles penetrate,some of the microneedles cannot penetrate the skin completely.Therefore,the drug distribution of the microneedle needs to be improved to improve the drug delivery efficiency.This research explored the type and concentration of the matrix material of the microneedles and the preparation method.The drug distribution of the microneedle prepared by the 25wt% polyvinyl alcohol (PVA) solution is more ideal.The hyaluronic acid (HA) has a poor ability to concentrate the drug distribution,the drug is easier to distribute on the microneedle base part.Using PVA to assist HA in the preparation of microneedles can achieve a more suitable drug distribution effect.The use of computer simulation analysis is consistent with the experimental results,which shows that it is feasible to use computer simulation to assist the basic research of drug delivery in microneedles.This research prepared antimicrobial laser-driven microneedles,which has good antibacterial effects.The antimicrobial material of this microneedle is a porous nanocarbon material.With the near-infrared laser as the driving force,the material converts light into heat to accelerate the dissolution of microneedles.The microneedles prepared with PVA as the matrix material have uniform drug distribution and better microneedle mechanical properties.The release effect on gelatin simulated skin is outstanding.Whether in vivo or in vitro,antimicrobial microneedles can achieve the purpose of effective antibacterial and no hidden cysts were found deep in the healing skin.The proposal of this new antibacterial method not only provides a new idea for the direction of microneedle loading drugs,but also provides a new bridge for the use of antibacterial drugs.
