Cancer has a high fatality rate and is still a serious health problem plaguing human beings.In recent years,a lot of money and manpower have been invested in the research of cancer treatment.Due to the complexity and diversity of tumors,the current clinical treatment methods are not ideal.Therefore,the research on cancer treatment has gradually shifted from monotherapy to combination therapy,and it is urgent to design a more efficient and reasonable platform for cancer treatment.In this study,lanretide acetate was used as a template to design and synthesize silver nanoscale cubes.In combination with photothermal therapy,starvation therapy,chemotherapy and other treatment methods,drug accumulation at tumor sites was improved by coating tumor cell membrane,side effects were reduced,and its anti-tumor performance was studied.The main research contents are as follows:1.The silver nano-cube was successfully prepared by biomimetic mineralization based on Lan.The synthesis mechanism of AgNCs was analyzed by TEM characterization of the solution at each step of synthesis.The effect of incubation time on the morphology of the synthesized particles was investigated to prove the guiding effect of Lan on crystal synthesis.Finally,AgNCs with good regular morphology and dispersion was prepared.The average particle size of AgNCs was 86 nm.UV-vis spectra showed that the prepared AgNCs had good photothermal conversion potential.2.Ag-G/C@M was designed and synthesized based on tumor targeting,cascade biocatalysis and photothermal coordination.TEM,UV-vis spectra,EDS energy spectrum,Zeta potential and other characterization demonstrated the successful preparation of Ag-G/C@M nanoparticles.The average particle size of Ag-G/C@M was 103 nm and the photothermal conversion efficiency was 26.2%.In vivo and in vitro experiments proved the homologous targeting effect of tumor cell membrane,and also proved that Ag-G/C@M could effectively inhibit tumor growth without obvious side effects.3.With PDA as the connector,GOx and TPZ were combined with PDA throughπ-πstacking and covalent bonding.Then tumor cell membrane was wrapped on the surface of nanoparticles by liposome extruder.While cutting off the internal glucose supply of tumor cells,GOx aggravates hypoxia at the tumor site and induces reactive activation of chemotherapy drug TPZ.The H2O2 produced can decompose Ag@PDA/GOx/TPZ@M,effectively reducing the side effects of nanoplatform while ensuring the treatment effect.
