In recent years,safety incidents related to food additives have frequently appeared in the public's field of view,which has aroused consumers' concerns about food safety;at the same time,consumers' dietary concepts of food and clothing have also been discovered from the past way to the current green health.Among them,the Chinese government and laws have expressly banned the use of additives that endanger human health,but there are still some food producers who seek to gain personal benefit by adding poppy shells to chafing dish condiments.In this study,the active ingredient of poppy shell-morphine is used as the test object,which is different from traditional chromatography,spectroscopy and electrophoresis.Immunolateral chromatography test strip methods for rapid detection of the main component of poppy shell is established to ensure humans health and social stability.The establishment of the method for the detection of morphine by colloidal gold nanoparticles is based on the specific recognition principle of antigen and antibody,the immune competition mechanism and the immunolateral chromatography.When the target substance is present in the sample,morphine is preferentially combined with the gold nanoparticles coupling with anti-morphine antibody,resulting in weakened or disappeared color development of the detection line.After simple pretreatment,under the optimal experimental conditions,morphine detection could be realized within 10 min,the detection limit of naked eye is 5 ppb,and the standard curve is linear in the range of 0.1-5 ppb.On the basis of the traditional strip,the semi-quantitative determination of morphine in the chafing dish condiments is realized through the different concentration of two detection lines.Under the optimal experimental conditions,morphine could be detected within 10 min.The detection limit of the naked eye is 5 ppb,which shows different good linearity in the range of 0.1-5 ppb and 1-30 ppb.By changing the selection of nanoparticles in the strips,fluorescent microspheres are used instead of gold nanoparticles,so that the binding mode of nanoparticles to morphine monoclonal antibody is changed from electrostatic adsorption to more stable chemical interaction.Under the best experimental conditions,morphine detection could be completed within10 min,and the detection limit of the naked eye is 0.5 ppb,which was 10 times more sensitive than the traditional method.The linear range of the standard curve was0.001-0.5 ppb.These results strongly prove that these methods have strong specificity and high sensitivity,and can be successfully applied to the field screening of the main component of poppy shells in chafing dish condiments.
