With the economic growth,the demand for fresh aquatic products has gradually increased,and the long-distance transportation of aquatic products has received much attention.In order to reduce the metabolism and stress response of aquatic products,fish anesthetics are commonly used for anesthesia transport preservation.Propofol is a new type of anesthetic in the preservation of aquatic products.There are a few reports on the residual amount,toxicity and harm to human body of propofol in aquatic products.What’s worse,there are no detection methods and limit standards for propofol anesthetic residues in aquatic products.It is necessary to establish a method for determining the residual amount of propofol anesthetic.In this paper,the prawn,cockroach and grouper were chosen as experiment objects.After pretreatment of samples,three detection methods were established for the residual amount of propofol anesthetic in aquatic products,including high performance liquid chromatography(HPLC),gas chromatography(GC)and gas chromatography-mass spectrometry(GC-MS)methods.The main research contents and results of this paper are as follows:1.Establishment of HPLC method for determination of propofol anesthetic residues in aquatic productsA method was established for the determination of propofol anesthetic residues in aquatic products by HPLC.The sample was extracted by acetonitrile and then purified by PRi ME HLB column(acetonitrile-water as mobile phase).The content of propofol was quantified by internal standard method.The results show that it has a good linear relationship in the range of 0.02 μg/m L to 1.00 μg/m L,and the correlation coefficient is R2=0.9992.When the concentration of propofol added in the sample is 0.20 mg/kg~1.00 mg/kg,the standard recovery is 82.82%~105.56%.The relative standard deviation is 1.92%~8.25%,and the limit of quantitation(LOQ)of the method is 0.10 mg/kg.The detection method with HPLC is suitable for detecting the residual amount of propofol anesthetic in aquatic products due to its rapid pretreatment,high precision and high recovery rate.2.Establishment of GC method for determination of propofol anesthetic residues in aquatic productsA method was established for the determination of propofol anesthetic residues in aquatic products by GC.The samples were extracted by ethyl acetate and then purified by EMR-Lipid combined with EMR-Polish column.The content of propofol was by internal standard method.The results show that it has a good linear relationship in the range of 0.40 μg/m L~10.00 μg/m L,and the correlation coefficient R2=0.9999.When the concentration of propofol added in the sample is 1.00 mg/kg~10.00 mg/kg,the standard recovery is 83.75%~102.18%.The relative standard deviation is1.54%~7.39%,and the limit of quantification(LOQ)of the method is 0.10 mg/kg.The detection method with GC is suitable for detecting the residual amount of propofol anesthetic in aquatic products due to its high aquracy and the less use of organic reagents.3.Establishment of GC-MS method for determination of propofol anesthetic residues in aquatic productsA method was established for the determination of propofol anesthetic residues in aquatic products by GC-MS.The sample was extracted by acetonitrile and then purified by Bond Elut plexa column,selective ion monitoring mode(SIM)and EI source.The content of propofol was by internal standard method..The results show that it has a good linear relationship in the range of 5.0 μg / L ~ 500.0 μg / L,and the correlation coefficient R2 = 0.9990.When the concentration of propofol added in the sample is 10.00 μg/kg~500.00 μg/kg,the standard recoveries is 86.93%~115.82%.The relative standard deviation is 1.61%~7.49%,and the limit of quantitation(LOQ)of the method is 10.0 μg/kg.It is cleaned by Bond Elut plexa column,and the sample pretreatment has good purification effect;the detection result has lower limit of quantification,which is a better detection method for propofol anesthetic residue detection in aquatic products.
