Ge(Germanium)is a very important element in vivo of human and animals,.As we know,germanium exists in nature in the form of inorganic germanium and organic germanium,Besides,the inorganic germanium artificially added to crops can be converted into organic germanium through crop enrichment and then enter the food chain to achieve the purpose of safe use by the human body.Barley seedlings are rich in protein,vitamins,minerals and other nutrients,therefore,it is a natural health food for humans and a high-quality livestock and poultry feed.In summary,this experiment takes the interaction between germanium and barley seedlings as the research direction,barley seedlings were cultivated with different cultivation methods(blank control group,adding germanium solution at soaking barley seeds stage,adding germanium solution at malted barley stage,and adding germanium solution at the whole process)and different concentrations(blank、5 mg/L、10 mg/L、15 mg/L、20 mg/L、30 mg/L)of germanium solution,to investigate the effects of different germanium application methods and different concentrations of germanium application on the growth of barley seedlings and the conversion efficiency of barley seedlings to inorganic germanium in the process of conversion of inorganic germanium into organic germanium through the enrichment of barley seedlings.Collecting four groups of barley seedlings with better growth and no toxicity including blank control,adding germanium solution of 5 mg/L at malted barley stage,5mg/L of germanium solution at soaking barley seeds stage and 10 mg/L of germanium solution at soaking barley seeds stage.By measuring the moisture content of barley seedlings,it was found that there was no significant difference in the moisture content of barley seedlings with different cultivation methods and different germanium-rich concentrations;However,as the germanium solution was added during the cultivation process,the ash content of the barley seedlings increased significantly by absorbing and enriching germanium in the culture solution;After comparing the barley plants treated with germanium solution with the blank control group,it was found that germanium can regulate soluble polysaccharides and soluble proteins in barley plants,and the order of regulation of soluble polysaccharides and soluble proteins is as follows: 10 mg/L(adding germanium solution at soaking barley seeds stage)> 5 mg/L(adding germanium solutionat soaking barley seeds stage)> 5 mg/L(adding germanium solution at malted barley stage)> Blank control group.The organic germanium and inorganic germanium in the above four groups of healthy and non-toxic barley seedlings were separated by dialysis bag method,and the content of total germanium and organic germanium in the wheat seedlings was determined by inductively coupled plasma emission spectrometry(ICP).After comparing the growth of wheat seedlings,impacts on other nutrients in plants and the conversion efficiency of organic germanium,it was found that the group of adding germanium solution in the concentration of 10 mg/L at soaking barley seeds stage showed a higher conversion rate of organic germanium and better growth,and the content of organic germanium in the plant is 29 times that of the blank control,it can be seen as the best wheat-germanium-rich group.The effects of different concentrations of germanium-rich barley seedlings on the proliferation of macrophages was determined using MMT for the purpose of exploring the auxiliary effect on cell activity and mechanism of germanium-rich barley seedlings.Four different concentrations(0.05 g/mL,0.1 g/mL,0.15 g/mL,0.2 g/mL)gradients were prepared respectively using germanium-rich barley seedling juice cultivated with 10 mg/L of germanium solution at soaking barley seeds stage.The test results showed that the proliferation rate of macrophage increased first and then decreased with the increasement of the concentration of germanium-rich barley seedling juice and peaked at the concentration of 0.15 g/mL,which proves that it has the greatest effect on the proliferation rate of macrophages in this experiment.
