Cordyceps sinensis is a complex of Cordyceps sinensis and bat larvae,and it is one of the three traditional valuable medicinal herbs.Cordyceps sinensis has various effects,such as enhancing immunity,anti-tumor and anti-fatigue.However,due to the shortage of medicine sources and the high cost,people have carried out a lot of research on the mycelium obtained by fermentation of the fungus isolated from Cordyceps sinensis.Isaria felina,as a new fungus isolated from Cordyceps sinensis,has good medicinal prospects.Therefore,the immune regulation function of IFMP and its isolated polysaccharides was studied in this work;The main chemical componenets of alcohol solubility of IFMP were analyzed and identified by HPLC-Q-TOFMS.The specific research contents and results are as follows:(1)Study on the immune activity of IFMPTo investigate the effects of IFMP on immune function in mice with immunosuppression induced by cyclophosphamide.The kunming mice were randomly divided into control group,model group,positive group(lentinan,200 mg·kg-1),low,medium,and high dose groups of IFMP(100,200,400 mg·kg-1).Except for the control group,the other five groups were intraperitoneally injected with cyclophosphamide to establish an immunosuppression mouse model.The drug was administered once a day for a total of 10 d.Then the body mass growth,liver index,thymus index,spleen index and blood routine indexes were examined;The carbon profile method and the delayed type hypersensitivity(DTH)was used to determine the non-specific immune function and the cellular immune function in mice;The content of TNF-α in serum of mice was determined by ELISA.The results showed that the detection indicators of the model group mice were significantly reduced(P<0.05).Compared with the model group,spleen index,white blood cells,red blood cells and other blood routine indexes,phagocytic index,plantar thickness difference and TNF-α content in each dose group of IFMP were significantly increased(P<0.05).In conclusion,IFMP has immunoprotective effects on immunocompromised mice induced by cyclophosphamide.(2)Study on extraction and immune activity of IFMPPIFMPP was extracted by water extraction and alcohol precipitation method.We observed the immunomodulatory effects of IFMPP by measuring the body weight increase value,organ index,blood routine index and lysozyme,IL-2,TNF-α,Ig G content in the serum of mice,and compared with the effects of the middle dose group of IFMPP and IFMP.The results showed that IFMPP could significantly increase the spleen index,thymus index,blood routine index,and lysozyme,IL-2,TNF-α,and Ig G levels in immunosuppressed mice(P<0.05)and the medium dose group of IFMPP had a better effect than the middle dose group of IFMP.In short,IFMP could promoted the immune function of the immunocompromised mice induced by cyclophosphamide.(3)Analysis and identification of main chemical components of IFMPBased on optimizing the conditions for the separation of alcohol extracts of IFMP by HPLC,this work determined the alcohol extracts of IFMP by HPLC-Q-TOF-MS and a total of 36 components were identified,including: nucleosides(18 species),amino acids(15 species),sterols(1 species)and glycosides(2 species),which collected data by positive and negative ion scanning mode,used the functions of finding and identifying compounds in the mass spectrometry workstation and combined the compound information provided by the database and the fragmentation law of secondary fragments in the literature.The results provided scientific basis for basic research,quality evaluation,active ingredient selection of IFMP,and laid a foundation for its development and utilization. |
