Both Monascus and Auricularia auricula are important medicinal and edible homologous fungus.Red koji is a kind of red rice grain fermented by Monascus purpureus,which takes rice as raw material.During the fermentation process,it can produce a variety of metabolites.It has the functions of reducing blood fat and pressure,anti cardiovascular disease,preventing atherosclerosis,etc.Auricularia auricula is a kind of saprophytic fungus.Its fruiting body contains polysaccharides and other active ingredients,which has the functions of antithrombotic,hypolipidemic and antitumor.In this project,the fermentation technology of Monascus solid-state fermentation of Auricularia auricula materials was established by using modern microbial solid-state fermentation technology.Taking Zhibituo,Auricularia auricula powder,Monascus powder,Auricularia auricula and Monascus powder as the control,the animal model of hyperlipidemia was used to evaluate the effect and safety of reducing blood lipid of fermentation powder of Auricularia auricula Monascus fermentation powder,and the forms of granules and capsules were developed.Meanwhile,the quality standard system of products was established.The specific research methods,results and conclusions are as follows:(1)Solid state fermentation of Auricularia auricula materials by four kinds of Monascus.The content of polysaccharide,reducing sugar,protein,lovastatin,citrinin,Monascus pigment and the antioxidant activity of Monascus pigment were used as indexes to screen Monascus strains with high pigment,high lovastatin and low citrinin.The results showed that the content of lovastatin was the highest in the fermentation product of Monascus M.h2019,reached 1724.19 μg/g,the content of Monascus pigment was the highest,was 2.74 U/ml,and the monascus pigment produced by this strain has high antioxidant activity,citrinin content was also lower than the national standard limit,so M.h2019 was selected as the best strain for the subsequent solid-state fermentation of Auricularia auricula Monascus.(2)In order to optimize the solid-state fermentation process of Monascus M.h2019 and Auricularia auricula,single factor experiment,Plackett-Burman experiment design,steepest climbing experiment design and Box-Behnken experiment design were carried out with Monascus pigment content in fermentation product as index.The best process is obtained by statistical optimization: raw material ratio(rice: agaric,g/g)= 4:1,fermentation time 18.61 d,rice cooking for 35 min,loading amount 22.36 g/250 mL,material liquid ratio(fermentation raw material: water,g/mL)= 1:2,inoculation amount of Monascus 13.40 mL,fermentation temperature 30 ℃,water addition amount 8.69%,water addition time 7 d.(3)In vivo test of hyperlipidemia model mice to evaluate the effect of Auricularia auricula Monascus on hypolipidemic.The results showed that Auricularia auricula Monascus could control the weight growth of high-fat mice,significantly reduce the organ index of liver and kidney in mice,the content of total cholesterol(TC)and triglyceride(TG),the content of low-density lipoprotein(LDL-C)and the content of high-density lipoprotein(HDL-C).All the efficacy indexes are far superior to the self-designed Auricularia auricula powder group,Monascus powder group,Auricularia auricula powder and Monascus powder compatibility group,and the existing functional foods for reducing blood lipid in the market.Therefore,after solid-state fermentation,Auricularia auricula Monascus has a better effect of hypolipidemic,and can be used for the development of functional foods of hypolipidemic.(4)The granules and capsules functional foods of Auricularia auricula Monascus were prepared by adding diluent,adhesive,flavoring agent and wetting agent.And carry out product quality inspection in accordance with national standards.The results showed that the particle size,water content,loss on drying,loading,loading difference,lovastatin content,citrinin content and disintegration time reached the corresponding product quality standards,and Auricularia auricula Monascus granules and capsules were qualified products.
