Porcine infectious pleuropneumonia(PCP)is a contact-prone and highly lethal respiratory infection caused by Actinobacillus pleuropneumoniae(APP).The cross-protection between various APP types is not strong,and there are also a large number of drug-resistant strains.In particular,in recent years,affected pigs have been infected with APP and often have mixed infections with other diseases,such as Swine fever,Porcine reproductive and respiratory syndrome,and Porcine circovirus disease,thus increasing the difficulty of treatment.Therefore,it is necessary to find safe,reliable and inexpensive new drugs.In view of the problems faced by PCP,according to the theory of traditional Chinese medicine and veterinary medicine,the characteristics of traditional Chinese medicine and the development mechanism of PCP,traditional Chinese medicine monomers(Phillyrin,Chlorogenic acid,TraxasterolL,Astilbin,Luteolin)and Chinese herbal compound(Huangren Tang,Shenghuang Decoction,Lianjin San Boil Liquid)were used to treat mice infected with APP.The experimental results showed that the clinical symptoms of mice treated with Phillyrin and Huangren Decoction were significantly improved(p<0.01),the lung index was significantly lower(p<0.01),and the survival rate was the highest,which indicating that Phillyrin and Huang Ren Tang had a good therapeutic effect on APP infected mice.To further investigate the therapeutic effect of Phillyrin and Huang Ren Tang on experimentally infected APP mice,we established a mouse infection model.ICR mice were randomly divided into 4 groups:normal control group,infection control group,Phillyrin treatment group,and Huangren Tang treatment group.The mice were infected with APP(2×10~8 CFU)in the nasal cavity.Two hours later,the phillyrin-treated group(20 mg/kg)and the Huangren Decoction-treated group(10 g/kg)were administered orally separately.The normal control group and the infection control group were both administered,which were poured in the same volume of 0.9%sodium chloride solution.They were gave medicine once every 12 h,and it lasted for3 days.The experimental results showed that the clinical symptoms of mice treated with Phillyrin and Huangren Decoction were significantly improved(p<0.01),the number of inflammatory cells in blood and BALF was significantly reduced(p<0.05),TNF-?,IL-6 The level of IL-1?was significantly decreased(p<0.05),and the pathological changes of the lung tissue were significantly reduced,which indicating that Phillyrin and Huang Ren Tang have a better therapeutic effect on APP-infected mice.In order to further verify the clinical efficacy of Phillyrin and Huang Ren Tang,we used 35-day piglets to study the therapeutic effects of APP-infected piglets.First,an artificial infection model of Porcine infectious pleuropneumonia was established by intranasal inoculation of APP.Thirty-nine piglets were divided into 5 groups:normal control group,infection control group,enrofloxacin treatment group,Phillyrin treatment group,and Huang Ren Tang Treatment group.Six hours later from the infection,the enrofloxacin-treated group,the Phillyrin-treated group,and the Huangren Tang-treated group were administered and treated twice a day,which lasted for 5 days.The experimental results showed that the Huangren Tang treatment group was 100%,which was the same as that of the enrofloxacin treatment group.At the same time,the weight gain rate of the Huangren Tang treatment group and the enrofloxacin treatment group was significantly higher than that of the infection control group(p<0.01),while there was no significant difference in the weight gain rate of the Huangren Tang treatment group and the enrofloxacin treatment group.(p>0.05).In summary,Phillyrin and Huangren Decoction were used for 5 days,which had a good therapeutic effect on PCP in piglets,which laid a foundation for further clinical application and could be used as a treatment for PCP.Moreover,they were safe,reliable and inexpensive optional drugs.
