The unique geographical environment has created the genetic specificity of Auricularia,but there are no reports on the collection,classification,identification and domestication of wild Auricularia in AfricaIn this study,the wild strains were isolated,Combining the antagonistic test of different strains,traditional morphological methods and molecular methods,the samples of Auricularia were identified.The high-yield and high-quality strains were selected through biological characteristics test,domestication cultivation and agronomic traits analysis.The aim is to collect,evaluate and domesticate wild Auricularia auricula germplasm resources in Africa for popularization and utilization.The results are as follows:1.Morphological identification and microscopic observation showed that there were significant differences in macro-structural and micro-structural characteristics of 9 strains.At the same time,combined with molecular phylogenetic analysis,it was found that strains BL1,JL2,JL4,JL7 and JL8 belonged to Auricularia cornea.LJ9 is a reference species,Auricularia cf:novozealandica.ZLJ10 is Auricularia novozealandica,ZL5 is a reference species,Auricularia cf: cornea.BL6 is a reference species,Auricularia cf: novozealandica,JL2,JL4,JL7 and JL8 are new record species in Zambia.ZL5 is a new record of the hairy fungus in the Zambian country.BL6 it is a new record species in Benin.ZLJ9 and ZLJ10 are new record species in Zambia,A total of 4 new records were found.2.Experiments on the biological characteristics of strain carbon source,nitrogen source,pH and temperature: BL1,JL2,JL4,JL7 and JL8.Sucrose is the most suitable carbon source for ZL5,BL6,ZLJ10 and ZLJ9.The best nitrogen sources of BL1,ZL5 and BL6 are yeast extract powder.The best nitrogen sources of JL4,JL7,JL8 and ZLJ10 are beef paste.The optimum nitrogen source for JL2 and ZLJ9 is peptone;The optimum pH of BL1,JL2,BL6 and ZLJ10 is 6.The optimum pH of ZLJ9 is 5.The optimum pH of JL4 is 9.The optimum pH of JL8 is8.The optimum pH of JL 7 and ZL5 is 7.The optimum mycelial growth temperatureof strain JL2 and strain ZLJ9 was 25?,and the others were 30?.Among them,the temperature has the most significant difference in the growth of hyphae.3.The dominant order of maturation time,germination rate,biological conversion rate,yield and ear thickness of fruiting bodies among 9 strains were analyzed comprehensively.Combining with ear type of strains,excellent strains BL1,JL8 and BL6 with commercial characteristics were screened.The above results indicated that four new records were found in three countries in the excavation of germplasm resources in Africa.The biological characteristics test showed that the mycelial growth of each strain was most suitable for carbon source,nitrogen source,pH and temperature,among which temperature had the most influence.Significantly;for the first time,systematically domesticated cultivation of African fungus germplasm resources;three high-yield,high-temperature,disease-resistant and excellent economic fungus new strains were screened,which had higher input-output ratio than domestic ones.Moreover,this study responded to the national “One Belt,One Road” policy call for assistance,and provided the foundation for the establishment of the “ One District,One Management,Five Warehouses” concept of the internationally renowned biologists China Engineering Center Li Yu.
